Einleitung: Herz-Kreislauferkrankungen als Folge einer arteriosklerotischen Veränderung der Blutgefäße werden durch Risikofaktoren wie Zigarettenrauchen, Übergewicht, Bluthochdruck, Diabetes mellitus und Bewegungsmangel begünstigt. Daneben werden die Konzentrationen im Blut an Serumlipiden, antioxidativ wirksamen Substanzen, Gerinnungsparametern oder ?neueren? Risikofaktoren wie z.B. C-reaktives Protein, Lipoprotein (a) oder Homocystein ins Feld geführt. Epidemiologische Studien legen nahe, dass moderater Alkoholkonsum das kardiovaskuläre Risiko senkt. Dies wird hauptsächlich einem Anstieg des HDL-Cholesterins im Blut zugeschrieben. Methoden: In der vorliegenden Studie wurde bei einem Probandenkollektiv von 284 gesunden Männern mit einem Alter zwischen 23 und 66 Jahren untersucht, ob Alkoholkonsum die Konzentration der vorangehend genannten kardiovaskulären Risikofaktoren beeinflusst. Zusätzlich wurde geprüft, ob ein Zusammenhang zwischen dem Alkoholkonsum und der Konzentration einiger Vitamine und von Substanzen besteht, die als protektive Faktoren für die Entwicklung kardiovaskulärer Erkrankungen angesehen werden. Im Vergleich zu bisher publizierten Studien mit ähnlicher Fragestellung unterschied sich die eigene Untersuchung zum einen durch die deutlich größere Zahl parallel gemessener Substanzen im Blut und zum anderen dadurch, dass die durchschnittliche tägliche Aufnahme von Makro- und Mikronährstoffen sowie der Konsum weiterer Genussmittel und anthropometrische Daten sorgfältig mit Hilfe eines computerunterstützten, validierten Ernährungsanamneseprogramms erhoben ermittelt und bei der Auswertung der Daten berücksichtigt wurden. Ausgehend vom durchschnittlichen täglichen Alkoholkonsum wurden die Probanden in drei Gruppen unterteilt: Gr. 1 (0-5 g), Gr. 2 (5,1-30 g) und Gr. 3 (30,1-70 g). Zur Prüfung, ob die Art des konsumierten alkoholhaltigen Getränks die untersuchten Messgrößen beeinflusst, wurden die Gruppen 2 und 3 nochmals unterteilt in ?Biertrinker Gr. 2+3? (> 80 % Bier), ?Weintrinker Gr. 2+3? (> 80 % Wein) und Konsumenten ohne Bevorzugung eines bestimmten alkoholhaltigen Getränks. Ergebnisse: Zwischen den nach Höhe und Art des Alkoholkonsums resultierenden Gruppen ergaben sich bezüglich der Hauptnährstoffe, der Energiezufuhr und des -bedarfs keine signifikanten Unterschiede. Bei den Markern für regelmäßigen, stärkeren Alkoholkonsum (g-GT und MCV) fand sich eine positive, signifikante Korrelation mit der Höhe der Alkoholaufnahme. Bei der großen Anzahl anderer klinisch-chemischer Laborparameter konnte kein Zusammenhang mit Höhe oder Art des Alkoholkonsums errechnet werden. Damit sind die Gruppen im Hinblick auf diese Laborparameter als vergleichbar anzusehen. Es gab ferner keinen Hinweis auf das Vorliegen leichterer Erkrankungen, welche die Ergebnisse der Lipid- und Vitaminmessungen beeinflussen könnten. Antioxidativ wirksame Substanzen im Blut:
Die Zufuhr an Vitamin B2, B6, B12, Folsäure, Retinol, ß-Carotin und anderer Carotinoide war in den Gruppen 1-3 vergleichbar. Die Probanden der Gr. 1 hatten eine höhere Zufuhr der Vitamine C, B1 und E, die ?Biertrinker Gr. 2+3? eine höhere Zufuhr an Vitamin B2, B6 und Folsäure. Der Vitamin B2-Status (EGRAC) war bei der Gr. 3 schlechter (vs. Gr. 1 bzw. Gr. 2). Umgekehrt war der Vitamin B6-Status bei der Gruppe 3 besser sowie die Konzentration von im Plasma frei vorliegendem Pyridoxalphosphat höher als bei Gr. 1. Die ?Weintrinker Gr. 2+3? hatten einen signifikant schlechteren Vitamin B6-Status (vs. ?Mischtrinker Gr. 2+3?, Tendenz: ?Biertrinker Gr. 2+3?). Die Gr. 3 hatte einen niedrigeren Plasmaspiegel an ß-Carotin und ß-Cryptoxanthin im Vergleich zur Gr. 1. Der in der Literatur postulierte positive Einfluss von moderatem oder gesteigertem Alkoholkonsum lässt sich aufgrund der Ergebnisse nicht durch Verbesserungen des Vitaminstatus bzw. durch Erhöhung der Konzentration an Vitaminen im Blut erklären. Die in Bier enthaltenen Vitamine führten nur beim Vitamin B6 zu einer Verbesserung des Vitaminstatus, während bei Vitamin B2 und Folsäure kein Einfluss zu errechnen war. Gesteigerter Alkoholkonsum (Gr. 3) brachte trotz vergleichbaren Zufuhrmengen bei Vitamin B2, ß-Carotin und ß-Cryptoxanthin eine Verschlechterung des Vitaminstatus bzw. eine Senkung der Plasmakonzentrationen im Vergleich zur Gr. 1 mit sich. Gerinnungsparameter, ?neuere? Risikofaktoren: Bei den Gerinnungsparametern Quick-Wert und Fibrinogen sowie den ?neueren? Risikofaktoren hochsensitives C-reaktives Protein und Homocystein war weder durch die Quantität noch durch die Art des Alkoholkonsums ein Einfluss zu erkennen. Lipoproteine: Die Konzentration an Gesamtcholesterin, Cholesterinester, Phospholipiden, Apolipoprotein A-1 und A-2 im Serum der Probanden aus Gr. 3 war höher. Moderater bzw. höherer Konsum von Alkohol führte zu einem signifikanten Anstieg der Cholesterinkonzentration in den High-density-Lipoproteinen (HDL) (inkl. Subfraktionen) ? unabhängig von der Art des alkoholhaltigen Getränks. Die Konzentration von Cholesterin in den Low- (LDL) (inkl. Subfraktionen), Very-low- (VLDL) und Intermediate-density-Lipoproteinen (IDL) im Blut wurde durch Alkoholkonsum nicht beeinflusst. Zusammensetzung der HDL: Die durch Alkoholkonsum induzierte Zunahme des HDL-Cholesterins war bei der Subfraktion HDL3 weniger stark ausgeprägt als bei den Subfraktionen HDL2b und HDL2a. Außerdem fand sich eine Veränderung der Zusammensetzung der HDL: Der Anteil an Phospholipiden nahm in stärkerem Maße zu als dies bei den anderen Komponenten der HDL der Fall war. Diese Zunahme steht möglicherweise in Zusammenhang mit dem Mechanismus der protektiven Wirkung der HDL auf Herz-Kreislauferkrankungen.
Background and aims of the study: Arteriosclerotic changes of blood vessels which contribute to coronary heart disease (CHD) and ischemic stroke are influenced by risk factors like cigarette smoking, overweight, hypertension, diabetes mellitus, missing physical exercise and nutritional factors, such as alcohol consumption. Beyond this, the concentrations of serum lipids, antioxidants, coagulation factors or other risk factors, such as C-reactive protein, and homocysteine are considered to be additional factors that indicate an enhanced or lowered risk of atherosclerosis. In this study we examined the effect of nutritional factors, in particular alcohol consumption, on various plasma components that are believed to play a role in the pathogenesis of atherosclerosis. Multiple epidemiologic studies suggest that moderate alcohol consumption reduces the mortality from cardiovascular diseases and that this effect is chiefly mediated by elevation of high-density lipoprotein (HDL). This cross-sectional study assessed the effect of moderate alcohol consumption and other life-style factors on the composition of HDL in healthy working males. An additional goal of the present study was to find out whether there is an association between alcohol consumption and the concentration of vitamins and cardio-protective substances. Methods: We included a collective of healthy men (n = 284, age 23-66 years), investigated with respect to cardiovascular risk factors. The average daily alcohol consumption, nutrient intake, smoking and other life-style factors was assessed by a computer based questionnaire. Group 1 (n = 62) comprised subjects with an average daily alcohol consumption of 0-5g, group 2 (n = 175): 5-30g, and group 3 (n = 47): 30-70g. In addition, the study design made it possible to subdivide groups 2 and 3 in a so called ?beer drinker group 2+3? (> 80 % beer), a ?wine drinker group 2+3? (> 80 % wine) and persons without preference of a certain alcoholic beverage. Results: The alcohol groups showed no significant differences in the nutritional profile (nutrients, energy intake, and metabolic rate). The markers for regular, higher alcohol consumption (g-GT and MCV) were positive correlated to the amount of alcohol consumption. There was no correlation between the great number of clinical laboratory parameters and the amount or kind of alcohol consumption. Thereby the groups are comparable in view of these laboratory parameters. Besides, there were no indications for the existence of diseases, which might influence blood lipid and vitamin concentrations. Antioxidative substances in the blood: Dietary assessment: The intake of vitamin B2, B6, B12, folic acid, retinol, ß-carotene and other carotenoids, as assessed by the computer interview was comparable in groups 1-3. The subjects in group 1 had a higher supply of the vitamins C, B1 and a-tocopherol, ?beer drinker Gr. 2+3? had a higher intake of vitamin B2, B6 and folic acid. Blood measurements: Antioxidative vitamins: The vitamin B2 status in erythrocytes (EGRAC) was lower in group 3 (vs. group 1 and 2). The plasma level of ß-carotene and ß-cryptoxanthin was lower in group 3 than in group 1. Vitamins that influence homocysteine metabolism (including homocysteine): Influence of beer and wine: The status of vitamin B6 and the concentration of free plasma pydridoxal phosphate in group 3 was significantly higher than in group 1. These results cannot explain the postulated positive influence of moderate or higher alcohol consumption through improvements of the vitamin status and the concentration of vitamins in the blood. The vitamins in beer improved the vitamin status only in case of vitamin B6, no effect was calculated in case of vitamin B2 and folic acid. Higher alcohol consumption (group 3) made the vitamin status respectively the plasma concentration of vitamin B2, ß-carotene and ß-cryptoxanthin lower compared with group 1 ? in spite of comparable supply. Coagulation factors, markers of inflammation: The coagulation factors prothrombin time and fibrinogen as well as the ?newer? risk factors C-reactive protein and homocysteine were not correlated to the amount or the kind of alcohol. Lipoproteins: The serum concentration of total cholesterol, cholesterol ester, phospholipids, apolipoprotein A-1 and A-2 was higher in group 3. Moderate and higher alcohol consumption raises the concentration of cholesterol in the high-density lipoproteins (HDL) (including subfractions) ? independent of the sort of alcoholic beverage. The concentration of cholesterol in the low- (LDL) (including subfractions), very-low (VLDL) and intermediate-density lipoproteins (IDL) in the blood was not influenced by alcohol consumption. Composition of HDL: The induced increase of HDL cholesterol was lower in the subfraction HDL3 as in the subfractions HDL2b und HDL2a. Besides we found qualitative changes of the HDL-components: the phospholipid component increased more than the other HDL-components. This phenomenon might play a beneficial role in the mechanism of atherosclerosis. Conclusions:
Vitamins: The changes of antioxidative vitamins and vitamins influencing homocysteine metabolism observed in persons with moderate and increased alcohol consumption do not explain the antiatherogenic effect of alcohol. On the other hand, our study confirmed a positive association of moderate alcohol consumption with HDL plasma levels ? independent of other nutritional factors. In addition, alcohol might induce qualitative alterations of HDL composition (more pronounced increased of HDL2 relative increase of the phospholipid component). The pathophysiological significance of this phenomenon remains unclear.

Synthesis of N-neterocycles via intramolecular reductive cyclizations of nitroalkenes

The current work describes a new triethyl phosphite mediated domino reaction of ω-nitro-alkenes which in one step cyclize to the corresponding saturated N-heterocycles. The one-step domino reaction described in this work includes a three-step reaction, a nitroso-ene reaction being coupled with two reduction reactions. A new C-N bond is formed and saturated N-heterocycles are synthesized. The microwave irradiation of the ω-nitroalkenes has been developed as an alternative protocol, having the advantage that the formation of byproducts can be suppressed and the reaction times reduced. The reductive cyclization reaction mediated by triethyl phosphite allows preparation of a series of saturated N-heterocycles such as substituted 3,4-dihydro-2H-1,4-benzoxazines, 1,2,3,4-tetrahydroquinoxalines and 1,2,3,4-tetrahydroquinolines.
The newly developed domino reaction can also occur under CO pressure using metal catalysts, allowing the formation of saturated N-heterocycles. Since the reductive cyclizations reported in this work are transition metal-catalyzed domino processes without precedent, they could be of significant interest to the field of synthetic chemistry and ?Life Sciences?.
Als Fazit lässt sich festhalten, dass die hier vorgestellte Arbeit eine neue durch Triethylphosphit vermittelte Dominoreaktion von ω-Nitroalkenen beschreibt, mit deren Hilfe die Cyclisierung zu entsprechenden gesättigten N-Heterocyclen in einem Schritt gelingt. Die einstufige Dominoreaktion umfasst insgesamt drei Reaktionen, und zwar eine Nitroso-En-Reaktion, die mit zwei Reduktionen gekoppelt ist. Eine neue C-N-Bindung wird gebildet und gesättigte N-Heterocyclen werden hergestellt. Die Bestrahlung der ω-Nitroalkene mit Mikrowellen wurde als ein alternatives Reaktionsprotokoll entwickelt. Es hat den Vorteil, dass die Bildung von Nebenprodukten unterdrückt wird und die Reaktionszeiten deutlich verkürzt werden können. Die reduktive, durch Triethylphosphit vermittelte Cyclisierung erlaubt die Herstellung einer Reihe von gesättigten N-Heterocyclen wie substituierten 3,4-Dihydro-2H-1,4-benzoxazinen, 1,2,3,4-Tetrahydrochinoxalinen und 1,2,3,4-Tetrahydro-chinolinen.
Die neu entwickelte Dominoreaktion kann auch unter Druck in CO unter Einsatz von Übergangsmetallkatalysatoren ablaufen, was zur Bildung von gesättigten Heterocyclen führt. Da die hier vorgestellten reduktiven Cyclisierungen bislang nicht untersuchte metall-katalysierte Dominoprozesse darstellen, könnten sie großes Interesse im Bereich der synthetischen Chemie und der ?Life Sciences? erregen.

Im Jahr 1993 isolierten Omura et al. aus dem Kulturüberstand von Aspergillus fumigatus FO 1289 eine neue Substanzklasse, die sich formal aus einem polyoxygenierten Sesquiterpen und einem 6-(3-pyridyl)-subsituierten α-Pyron ableiten ließ und deswegen Pyripyropene genannt wurden. Bis heute inhibieren Pyripyropene von allen getesteten Substanzen das Enzym Cholesterol-O-Acyltransferase (ACAT), das eine Schlüsselfunktion im Fettstoffwechsel beim Menschen ausübt, am wirkungsvollsten. Solche Verbindungen stellen einen vielversprechenden Ansatz bei der Behandlung von Krankheiten wie Arteriosklerose, die ihre Ursache in der Störung des Fettstoffwechsels haben, dar.
Anfänglich wurden Vorversuche zur verbesserten Totalsynthese von Pyripyropen E nach literaturbekannten Verfahren durchgeführt. Aus den großen Schwierigkeiten beim Aufbau der Schlüsselverbindungen wurden die folgenden Arbeitsschwerpunkte der vorliegenden Doktorarbeit festgelegt: a) selektive γ-Acylierung von 2-substituierten Acetessigsäureestern, b) effektive Umwandlung von β, δ-Diketocarbonsäuren in 4-Hydroxy-2H-pyran-2-one, c) optimierte Cyclisierung von epoxyolefin-substituierten 6-Pyridyl-4-hydroxy-2H-pyran-2-onen durch Variation der Parameter Lewis-Säuren, Lösungsmittel und Temperatur, d) Entwicklung einer neuen effektiven Methode für die Synthese von 6-substituierten 4-Hydroxy-2H-pyran-2-onen und anderer Heterocyclen und e) Anwendung der unter a) bis d) ausgearbeiteten Ergebnisse bei der effizienten Totalsynthese von drei Naturstoffen mit 4-Hydroxy-2H-pyran-2-on-Gerüst.
Das Cyclisierungssubstrat der Geranylreihe, das 3-[7-(2,3-Epoxy-2,3-dihydrogeranyl)]-6-(3-pyridyl)-4-hydroxy-pyran-2H-pyran-2-on, wurde in einer neunstufigen Synthese aus Geraniol mit 18 % Gesamtausbeute aufgebaut und dann in Cyclisierungsexperimenten mit zwölf verschiedenen Brønsted- bzw. Lewis-Säuren umgesetzt. Dabei wurden in flüssigem Schwefeldioxid abhängig von der Lewis-Säure bis zu vier Produkte isoliert: ein Pyrano[4,3-b]chromen-1-on (α-Pyron), ein Pyrano[2,3-b]chromen-4-on (γ-Pyron), ein 7-Oxa-bicyclo[2.2.1]hept-2-an und ein durch Epoxidöffnung entstandenes Diol, dessen Struktur durch Totalsynthese bewiesen wurde.
In der Farnesylreihe wurde das Cylisierungssubstrat analog desjenigen der Geranylreihe in einer neunstufigen Synthese mit 20 % Gesamtausbeute, ausgehend von (E,E)-Farnesol, aufgebaut und dann cyclisiert. Es wurde ein untrennbares Gemisch aus acht verschiedenen Isomeren erhalten.
Bei Iodcyclisierungen wurde 3-(7-Geranyl)-6-(3-pyridyl)-4-hydroxy-2H-pyran-2-on einmal ohne Base mit Iod in Acetonitril umgesetzt. In einem anderen Experiment wurde zuerst durch Deprotonierung mit Kaliumcarbonat das korrespondierende Enolatanion erzeugt, welches dann mit Iod zur Reaktion gebracht wurde. Ohne Base wurde das Iod-substituierte Pyrano[2,3-b]pyran-4-on-Derivat (γ-Pyron) gebildet, bei Umsetzung mit Iod nach Deprotonierung entstand dagegen eine Mischung aus zwei Iod-α-pyronen: einem Pyrano[4,3-b]pyran-5-on (endo-Produkt) und einem Furo[3,2-c]pyran-4-on (exo-Produkt).
Aus den gesammelten Erfahrungen beim Aufbau der Substrate für die Cyclisierungsexperimente in der Geranylserie wurde eine neue Synthese von 6-substituierten 4-Hydroxy-2H-pyran-2-onen entwickelt: das Hauptprinzip beruht auf der in situ Freisetzung der außerordentlichen empfindlichen 5-Hydroxy-3-oxo-pent-4-ensäuren aus ihren stabilen Biskalium-Salzen mit TFA bei tiefen Temperaturen, gefolgt von einer spontanen Lactonisierung dieser Säuren im Reaktionsmedium TFAA zu den korrespondierenden Pyronen. Die Effektivität dieses Verfahrens wurde an zwölf Beispielen eindrucksvoll demonstriert.
Das 5-Phenyl-substituierte Biskalium-Salz wurde außerdem zum effektiven Aufbau von Heterocyclen genutzt: es wurden jeweils ein Pyrazol und ein Isoxazol in hohen Ausbeuten synthetisiert.
Aus der neuen Pyronsynthese wurde die Totalsynthese von drei Naturstoffen mit 4-Hydroxy-2H-pyran-2-on-Gerüst abgeleitet:
1. Die aus Pseudomonas fluorescens isolierte Verbindung Sch-419560 wurde in einer vierstufigen Synthese, ausgehend von 2-Hexylacetessigsäureethylester, mit 62 % Gesamtausbeute hergestellt.
2. Das aus Garcinia conrauana Engl. (Guttiferae) isolierte Pyron 3?,3?-Dimethylallylconrauanalacton wurde, beginnend mit 2-Prenylacetessigsäureethylester, in einer fünfstufigen Synthese mit 64 % Gesamtausbeute hergestellt.
3. Abschließend konnte der aus Mimulus aurantiacus isolierte Naturstoff Aurantiacon in einer siebenstufigen Synthese, ausgehend von 2-Hydroxybuttersäure, mit 62 % Gesamtausbeute synthetisiert werden.
In 1993 Omura et al. isolated from the culture broth of Aspergillus fumigatus FO 1289 a new class of compounds that exhibit a polyoxygenated sesquiterpene and α-pyrone and therefore was called pyripyropenes. Up to now pyripyropenes represent the most potent inhibitors of the enzyme cholesterol-O-acyl transferase (ACAT) that plays a key role in human fat metabolism. For example, arteriosclerosis have its origin in a fat digestion disorder and therefore ACAT-inhibiting agents like pyripyropenes shows a new promising approach in the treatment of this cholesterol level depending diseases.
In the beginning preliminary studies were performed on improving the synthesis of pyripyropene E according to literature known procedures. From the big difficulties arriving from the synthesis of the key compounds this work focussed on: a) selective γ-acylation of 2-substituted aceto acetic esters, b) effective conversion of β, δ-diketo carboxylic acids into 4-hydroxy-2H-pyran-2-ones, c) optimized cyclization of epoxyolefine substituted 6-pyridyl-4-hydroxy-2H-pyran-2-ones by variation of different parameters like Lewis acids, solvents and temperature, d) design of a new efficient method for the synthesis of 6-substituted 4-hydroxy-2H-pyran-2-ones and related heterocycles, e) application of the in a) to d) elaborated results on the efficient total synthesis of three naturally occurring compounds with a 4-hydroxy-2H-pyran-2-on skeleton.
The cyclization substrate in the geranyl series, the 3-[7-(2,3-epoxy-2,3-dihydrogeranyl)]-6-(3-pyridyl)-4-hydroxy-pyran-2H-pyran-2-one, was synthesized in a nine step sequence starting from geraniol with 18 % overall yield. In the following experiments this epoxide was cyclized in liquid sulphur dioxide with twelve different Brønsted and Lewis acids. Dependent on the Lewis acid up to four products were isolated: a pyrano[4,3-b]chromen-1-one (α-pyrone), a pyrano[2,3-b]chromen-4-one (γ-pyrone), a 7-oxa-bicyclo[2.2.1]hept-2-ane and a diol generated from opening of the epoxide function. The structure of the diol was unambiguously determined by total synthesis.
In the farnesyl series the cyclization substrate was assembled in total analogy to that one of the geranyl series in a nine step sequence with 20 % overall yield starting from (E,E)-farnesol. After the cyclization experiments an inseparable mixture of eight different isomers were obtained.
During a series of iodocyclization experiments 3-(7-geranyl)-6-pyridyl-4-hydroxy-2H-pyran-one was reacted with iodine in acetonitrile without a base. In another experiment the corresponding enolate anion was generated by deprotonation with potassium carbonate and then was brought to reaction with iodine. Without base the iodo substituted pyrano[2,3-b]pyran-4-one derivative (γ-pyrone) was formed. The reaction of the enolate anion with iodine resulted in a mixture of two iodo substituted α-pyrones, a pyrano[4,3-b]pyran-5-one (endo product) and a furo[3,2-c]pyran-4-one (exo product).
From the experience that was gained by the assembly of the cyclization substrates in the geranyl series a new synthesis of 6-substituted 4-hydroxy-2H-pyran-2-ones was developed: The main principle was the in situ release of the extraordinary sensitive 5-hydroxy-3-oxo-pent-4-enoic acids through protonation of their stable bispotassium salts with TFA at low temperature, followed by spontaneous lactonization of these acids in the reaction media TFAA leading to the corresponding pyrones in high yields. The effectiveness of this procedure was impressively demonstrated in twelve examples.
The 5-phenyl-substituted bispotassium salt was used for the construction of heterocycles: one pyrazole and one isoxazole were synthesized in high yields.
Based on the new pyrone synthesis the total synthesis of three natural products with a 4-hydroxy-2H-pyran-2-one framework was carried out.
1. The compound Sch-419560 isolated from Pseudomonas fluorescens was synthesized in a four step sequence starting from ethyl 2-hexylacetoacetate with 62 % overall yield.
2. The α-pyrone 3?,3?-dimethylallylconrauanalactone isolated from Garcinia conrauana Engl. (Guttiferae) was synthesized via a four step sequence starting from ethyl 2-prenylacetoacetate with 64 % overall yield.
3. Finally, the natural product aurantiacone isolated from Mimulus aurantiacus was synthesized via a seven step sequence starting from 2-hydroxybutyric acid with 62 % overall yield.

The dissertation presents in its introduction the sources and process of food contaminations deriving from packaging materials. Subsequent legislative aspects, the analysis of food contact materials and contaminants in food are explained and examples therefore are given.

Die Dissertation erklärt in der Einleitung die Quellen und Mechanismen der Lebensmittelkontamination mittels Verpackungsmaterialien. Nachfolgend werden legislative Aspekte, die Analyse von Lebensmittelkontaktmaterialien und Kontaminanten in Lebensmitteln und aktuelle Beispiele hierfür behandelt.

Planar-chromatography (High-Performance Thin-Layer Chromatography, HPTLC) is a rapid and cost-effective offline separation method. Through advances in the automatization of each step the system reproducibility, from application and development to detection, has been improved. This makes planar-chromatography a highly reliable technique. HPTLC shows a couple of features that make it unique. There is great flexibility concerning application, development and detection that distinguishes HPTLC from other techniques. Especially the parallel development of up to 36 tracks per plate, the possibility of pre-chromatographic derivatization on the stationary phase, application volumes from nL up to mL, two-dimensional development, automated single or multiple development, and the multiple detection with different methods (UV, fluorescence, bioluminescence, etc.) have to be emphasized. A further advantage over column- (LC) and gas-chromatography (GC) is the single use of the stationary phase. This leads to a high tolerance towards sample matrix and allows for reducing sample preparation. Because of these aspects, planar-chromatography is an interesting tool for each analyst.
However, in the last years hyphenation with mass spectrometry (MS) did not make great advancements in comparison to HPLC and GC: thus, planar-chromatography became less attractive. Therefore an existing universal hyphenation (ChromeXtract by Dr. Luftmann), that was based upon a plunger for elution, was improved (publication 1). The original version of the plunger did not allow any elution from glass backed plates, since they broke easily under the pressure applied during clamping. It was difficult to adjust the pressure depending on the experience of the operator. Furthermore, solvent leakage was possible because of insufficient sealing of the cup-point. For a reproducible contact pressure that was independent from the experience of the operator, a commercial torque wrench was used for clamping of the plates. This guaranteed reproducible contact pressure. The installation of a small plastic buffer into the plunger ensured a slight kind of attenuation. This decreased the frequency of leakage from over 50 % to below 5 %.
An important criterion of applicability of this hyphenation is the repeatability of the extractions and thus the measurements. Thus, zones of xanthylethylcarbamat (XEC) and dansylpropanamid (DPA) were extracted after chromatographic development. Their specific masses were detected in positive ESI-mode. The relative standard deviation of the signal in single-ion-monitoring (SIM) mode was 18.6 % for XEC and 8.7 % for DPA. Linearity was given in the range of 10 to 200 ng/zone with a very good correlation coefficient (r > 0.9919). The limit of quantification at an S/N-ratio of 10 was calculated by means of the blank signal and amounted 52 and 160 pg/zone for XEC and DPA, respectively. Additionally, the influence of the elution solvent on the extraction of the HPTLC-plate and signal intensity was demonstrated with tests using different solvents.
The second publication addressed the application of planar-chromatography hyphenated with MS by means of the modified ChromeXtractor on the determination of acrylamide in drinking water. The strict limit within the EU of 0.1 mug/L until then was only controlled through costly methods that were almost exclusively based on GC-MS or LC-MS/MS after applying intensive clean-up procedures. Thus it was aimed to develop a low priced and rapid alternative method for routine analysis based on HPTLC. Therefore a pre-chromatographic in-situ derivatization of acrylamide with a fluorescence marker was used. The product was detected densitometrically after chromatographic separation. During development of the method, the mass of the reaction product was determined for analysis of the derivatization step. With the aid of the modified ChromeXtract the product could be directly extracted from the plate and transferred to MS. The exact mass proved that instead of the originally used fluorescence marker dansylhydrazine the dimethylaminonaphthaline(Dan)-sulfinic acid reacts with acrylamide. Consequently, dansulfinic acid was synthesized and used for derivatization.
To take advantage of the high tolerance of planar-chromatography towards various sample matrices, an approach was searched in order to skip sample preparation. However the necessity to use excess of reagent led to high background fluorescence. This allowed only a limit of detection of 20 mug/L. Thus, sample preparation and analyt enrichment was necessary to obtain a method able to control the maximum concentration. In accordance with DIN 38413-6 concerning determination of acrylamide in drinking water, activated carbon was used for analyte enrichment by means of solid phase extraction (SPE). An internal standard (dimethylacrylamide) was added prior sample preparation. The final extract was analysed as described. In spiked samples of drinking water, a 1000-fold lower limit of detection of 0.02 mug/L and a very good mean reproducibility across the whole system was shown, which suffices to control the maximum amount. A comparative study with measurements by LC-MS/MS revealed satisfactory correlation. Thus, for the first time a planar-chromatographic method for the determination of acrylamide at ultra-trace levels were presented.
The third publication addresses the application of the developed method on a very complex food matrix like coffee. Several publications reported problems during determination of acrylamide in coffee. Therefore the extremely high tolerance of planar-chromatography towards sample matrix effects was used, allowing for a shortened sample preparation. The idea of a rapid method was followed by the extraction of commercial coffee samples by means of accelerated solvent extraction (ASE). This allowed for higher throughput during sample preparation. To remove a part of the co extracted matrix, the whole ASE-extract was cleaned by SPE with activated carbon and evaporated to a defined volume. This represented a simplification of common multistage extraction methods and clean-up steps, that aim for complete removal of co extracted matrix prior injection into LC- or GC-systems. In accordance with determination of acrylamide in drinking water, the extract was derivatized in-situ with the fluorescence marker Dansulfinic acid and detected densitometrically after chromatographic separation. The concentration of acrylamide was quantified by means of parallel preparation of three standard additions. Systematic errors and the influence of the sample were corrected by the calibration within the matrix. The linearity of the calibration (between r = 0.9825 and 0.9995) were acceptable. Good values were reached for the limit of quantification (48 mug/kg) and repeatability (rsd 3 %). After method development the acrylamide concentration of commercial coffee samples was determined, showing results being consistent with literature findings. Thus the applicability of the newly developed method to complex food samples was demonstrated.
In summary, the present work shows the applicability of planar-chromatography hyphenated with mass spectrometry for sensitive determination of acrylamide. It was possible to quantify the analyte at ultra-trace levels using less instrumental effort and time than usual. Quantification in complex sample matrices was feasible in spite of a simplified sample preparation. These applications prove the relevance of planar-chromatography to solve current analytical problems.
Bei der Planar-Chromatographie (High-Performance Thin-Layer Chromatography, HPTLC) handelt es sich um eine schnelle und kosteneffektive offline Trenntechnik. Die Entwicklungen in der Automatisierung der einzelnen Schritte führten zu einer stetigen Verbesserung der Reproduzierbarkeit von der Auftragung über die Entwicklung bis hin zur Detektion. Dies macht die Planar-Chromatographie zu einer zuverlässigen Analysetechnik. Unter mehreren Gesichtspunkten ist die HPTLC eine besondere Technik. Ihre extreme Flexibilität in der Auftragung, Entwicklung und Detektion heben sie von anderen Techniken hervor. Die parallele Entwicklung von bis zu 36 Banden pro Platte, die Möglichkeit zur prächromatographischen Derivatisierung auf der stationären Phase, mögliche Auftragsvolumina von nL bis mL, zweidimensionale Entwicklung, automatisierte Einfach- und Mehrfachentwicklung und die mehrfache Detektion einer Platte mit verschiedenen Techniken (UV, Fluoreszenz, Biolumineszenz, etc.) sind dabei hervorzuheben. Ein weiterer Vorteil gegenüber der Säulen- (LC) oder Gas-Chromatographie (GC) ist die einmalige Benutzung der stationären Phase. Dies hat eine sehr hohe Toleranz gegenüber Probenmatrix und damit verbunden eine verringerte Probenaufarbeitung zur Folge. Dies alles macht die Planar-Chromatographie für den Analytiker zu einer interessanten Analysetechnik.
Jedoch machte in den letzten Jahren die Kopplung mit der Massenspektrometrie (MS) nicht den glei-chen Fortschritt, wie bei der HPLC und GC, wodurch die Planar-Chromatographie an Attraktivität verlor. Es wurde daher eine vorhandene universelle Kopplungsmöglichkeit (ChromeXtract von Dr. Luftmann), die auf einem Elutionsstempel basierte, verbessert (Publikation 1). Die ursprüngliche Version des Elutionsstempel liess keine Anwendung auf Glasplatten zu, da diese beim Einspannen schnell unter dem Anpressdruck des Stempels zerbrachen. Der Anpressdruck des Stempels liess sich nur schwer regulieren und war von der Erfahrung des Anwenders abhängig. Des Weiteren konnte ein Auslaufen des verwendeten Lösungsmittels durch unzureichende Abdichtung der Zone durch die Ringschneide auftreten. Um den Anpressdruck reproduzierbar und unabhängig vom Benutzer zu machen, wurde ein handelsüblicher Drehmomentschlüssel zum Einspannen der Platte verwendet. Dies garantierte einen reproduzierbaren Anpressdruck beim fixieren. Durch den Einbau eines Kunststoff-Puffers in den Stempel wurde ein leichter Ausgleich beim Anpressen gewährleistet. Dadurch konnte die Häufigkeit des Auslaufens von über 50 % auf unter 5 % gesenkt werden.
Ein wichtiges Kriterium für die Anwendbarkeit der Kopplungsmethode ist die Wiederholbarkeit der Extraktionen und damit auch der Messungen. Hierfür wurden mehrere Zonen der Substanzen Xanthylethylcarbamat (XEC) und Dansylpropanamid (DPA) nach ihrer chromatographischen Entwicklung extrahiert und deren spezifische Massen im positiven ESI-Modus detektiert. Die relative Standardabweichung des Single-Ion-Monitoring (SIM) Signals lag bei 18,6 % für XEC und 8,7 % für DPA. Die Linearität war über einen Bereich von 10 bis 200 ng/Zone bei sehr guten Korrelationskoeffizienten (r > 0.9919) gewährleistet. Anhand des Blindwertsignals konnten die Bestimmungsgrenzen bei einem Signal/Rausch-Verhältnis von 10 zu 52 und 160 pg/Zone für XEC und DPA berechnet werden. Zudem wurde der Einfluss des Elutionsmittels auf die Extraktion der HPTLC-Platte und die Signalintensität anhand von Versuchen mit verschiedenen Lösungsmitteln gezeigt.
Die zweite Publikation befasste sich mit der Anwendung der Planar-Chromatographie gekoppelt mit der MS mittels des modifizierten ChromeXtractors auf die Analyse von Acrylamid in Trinkwasser. Der in der EU geltende strenge Grenzwert von 0,1 mug/L konnte bisher nur von wenigen aufwändigen Bestimmungsmethoden, meist mittels GC-MS oder LC-MS/MS nach intensiver Probenaufbereitung, überprüft werden. Es sollte daher mit der HPTLC eine günstige und schnelle Alternativmethode für die Routine entwickelt werden. Hierfür wurde Acrylamid mit einem Fluoreszenzmarker prä-chromatographisch in-situ derivatisiert und nach chromatographischer Trennung densitometrisch detektiert. Bei der Entwicklung der Methode wurde zur Untersuchung des Derivatisierungsschrittes die Masse des Reaktionsproduktes bestimmt. Mittels modifiziertem ChromeXtract konnte dabei das Produkt direkt von der Platte extrahiert und ins MS geleitet werden. Die exakte Masse half dabei zu erkennen, dass statt des ursprünglich eingesetzten Fluoreszenzmarker Dansylhydrazin dessen Dimethylaminonaphthalin (Dan)-Sulfinsäure mit Acrylamid reagiert. Dies hatte zur Folge, dass Dansulfinsäure gezielt synthetisiert und eingesetzt wurde.
Um die hohe Matrixtoleranz der Planar-Chromatographie zu nutzen, wurde zunächst nach einem Ansatz gesucht, die Probenaufarbeitung zu überspringen. Die Notwendigkeit einen Überschuss des Reagenz für die direkte Umsetzung in der Wasserprobe zu verwenden, führte jedoch im Chromato-gramm zu einer starken Hinergrundfluoreszenz. Dies ermöglichte nur eine Nachweisgrenze von 20 mug/L. Um eine Methode zur Überprüfung des Grenzwertes zu erhalten, war daher eine vorherige Probenaufbereitung und Analytanreicherung nötig. Gemäss der DIN 38413-6 über die Untersuchung von Trinkwasser auf Acrylamid wurde der Analyt unter Verwendung eines internen Standards (Di-methylacrylamid) mittels Festphasenextraktion (Solid Phase Extraction, SPE) an Aktivkohle angereichert und der Extrakt wie beschrieben untersucht. Anhand von dotierten Trinkwasserproben konnte eine um den Faktor 1000 niedrigere Nachweisgrenze von 0,02 mug/L und eine sehr gute durchschnittliche Reproduzierbarkeit über das gesamte System gezeigt werden, was ausreicht den geltenden Grenzwert zu überprüfen. Vergleichsuntersuchungen mit HPLC-MS/MS-Messungen zeigten eine äusserst zufriedenstellende Korrelation. Insgesamt konnte zum ersten Mal eine planar-chromatographische Methode vorgestellt werden, die es ermöglicht, Acrylamid im Ultra-Spurenbereich zu bestimmen.
In der dritten Publikation wurde die entwickelte Bestimmungsmethode auf die sehr komplexe Lebensmittelmatrix Kaffee übertragen, denn es wurde in mehreren Publikationen hierbei von Problemen bei der Acrylamid-Bestimmung berichtet. Hierfür konnte die Matrixtoleranz der Planar-Chromatographie genutzt werden, die eine verkürzte Probenaufarbeitung ermöglichte. Der Ansatz einer schnellen Untersuchungsmethode wurde durch die Extraktion von kommerziell erhältlichen Kaffees mit Hilfe der beschleunigten Lösungsmittelextraktion (Accelerated Solvent Extraction, ASE) weiter verfolgt. Dadurch wurde ein grösserer Probendurchsatz bei der Aufbereitung möglich. Um einen Teil der mit-extrahierten Matrix zu entfernen, wurde der gesamte ASE-Probenextrakt einer SPE mit Aktivkohle unterzogen und anschliessend auf ein definiertes Volumen eingeengt. Dies stellte eine Vereinfachung zu den üblichen mehrstufigen Extraktions- und Aufreinigungsschritten dar, die eine weitestgehende Entfernung der coextrahierten Matrix vor der Injektion in ein HPLC- oder GC-System zum Ziel hatten. Der so vorbereitete Extrakt wurde analog der Bestimmungsmethode für Wasser in-situ mit dem Fluoreszenzmarker Dansulfinsäure umgesetzt und nach chromatographischer Auftrennung densitometrisch erfasst. Durch die parallele Aufarbeitung von drei Standardadditionen konnte der Acrylamidgehalt bestimmt werden. Die Kalibration in der Matrix korrigierte systematische Fehler und Einflüsse der Probe über die gesamte Methode. Die Linearitüt der Kalibrationen (zwischen r = 0.9825 und 0.9995) war akzeptabel. Die Bestimmungsgrenze und Wiederholbarkeit zeigten mit 48 mug/kg und 3 % relativer Standardabweichung gute Werte. Nach der Methodenentwicklung wurde von kommerziell erhältlichem Kaffee der Acrylamidgehalt bestimmt, wobei die erhaltenen Werte mit Angaben aus der Literatur übereinstimmten. Es wurde damit die Anwendbarkeit der neu entwickelten Methode auf schwierige Lebensmittel demonstriert.
Zusammengefasst zeigt die vorliegende Arbeit die Anwendbarkeit der Planar-Chromatographie in Verbindung mit der Massenspektrometrie für die empfindliche Bestimmung von Acrylamid. Es ist gelungen, mit einem Bruchteil des üblichen apparativen und zeitlichen Aufwands den Analyten zum einen bis hin zum Ultra-Spurenbereich zu quantifizieren. Des weiteren wurde trotz einer verkürzten Probenaufarbeitung die Quantifizierung in einer problematischen Matrix möglich. Diese Anwendungen stellen die Relevanz der Planar-Chromatographie für moderne analytische Fragestellungen unter Beweis.

In der vorliegenden Arbeit wurden neuartige Dominoreaktionen beschrieben, die auf Laccase-katalysierten Oxidation von Brenzcatechinen und Hydrochinonen zu den entsprechenden o- und p-Chinonen und sich daran anschließenden Umsetzungen mit 1,3-Dicarbonylen beruhen.
Im ersten Teil dieser Dissertation wurde ein effizienter Zugang zu 3,4-Dihydro-7,8-dihydroxy-2H-dibenzofuran-1-onen entwickelt, der sich die Laccase-initiierte Dominoreaktion zwischen Cyclohexan-1,3-dionen und Brenzcatechinen mit Luftsauerstoff als Oxidationsmittel Zunutze macht. Die Reaktionen konnten unter milden Reaktionsbedingungen und unter Vermeidung von toxischen Reagenzien durchgeführt werden. Die Produkte entstanden mit Ausbeuten zwischen 70 und 97% und mit hoher Reinheit. Nebenprodukte traten nicht auf. Die Struktur aller Verbindungen wurde zweifelsfrei durch NMR-spektroskopische Methoden aufgeklärt.
Im zweiten Teil der Arbeit wurden Laccase-initiierte Reaktionen zwischen Brenzcatechinen und heterocyclischen 1,3-Dicarbonylen (Pyridinone, Chinolinone, Thiocumarin) beschrieben, bei denen Benzofuropyridinone, Benzofurochinolinone und Thiocoumestane entstanden. Die Umsetzungen lassen sich einfach durchführen und liefern die Produkte regioselektiv und mit Ausbeuten zwischen 55 und 98%. Mit Barbitursäurederivaten beobachtete man ausschließlich die Bildung von Dispiropyrimidinonen. Die eindeutige und vollständige Strukturaufklärung aller Reaktionsprodukte gelang mittels NMR-spektroskopischer Methoden (HSQMBC und Band-selektives HMBC), sowie durch Röntgenstrukturanalyse.
Im dritten Teil der Arbeit wurden die Laccase-katalysierten Reaktionen von unterschiedlich substituierten Hydrochinonen mit cyclischen 1,3-Dicarbonylen untersucht. Die Reaktionen bieten einen hochselektiven Zugang zu Bisaddukten, bei denen die beiden 1,3-Dicarbonyl-Substituenten immer benachbart angeordnet sind. Eine Ausnahme ist die Umsetzung von Chlorhydrochinon mit 4-Hydroxy-cumarin, da hier ein Trisaddukt entsteht. Dieses Ergebnis ist vor allem deshalb bemerkenswert, weil bei der Umsetzung von Hydrochinonen mit 1,3-Dicarbonylen unter anderen Reaktionsbedingungen in der Regel die Bildung von Benzofuranderivaten beobachtet wurde. Die Strukturaufklärung wurde durch NMR-spektroskopische Methoden, wie die Spin-Muster Analyse der long-range gekoppelten C=O-Kohlenstoffatome und die 13C-Satelliten-Analyse in den 1H-NMR-Spektren durchgeführt.
Mit den hier entwickelten Dominoreaktionen gelingt die effiziente und selektive Darstellung einer ganzen Reihe von Heterocyclen sowie von substituierten p-Benzochinonen unter milden Reaktionsbedingungen. In den meisten Fällen lassen sich die Produkte in guten bis sehr guten Ausbeuten und mit hoher Reinheit isolieren. Für die Strukturaufklärung der Produkte wurde u. a. eine ganze Palette von NMR-Methoden herangezogen.
In the present work novel domino reactions have been described which are based on the laccase-catalyzed oxidation of catechols and hydroquinones to the corresponding o- and p-quinones and their subsequent reactions with 1,3-dicarbonyl compounds.
In the first part of this thesis an efficient approach to 3,4-dihydro-7,8-dihydroxy-2H-dibenzofuran-1-ones has been developed. The method includes a laccase-initiated domino reaction between cyclohexane-1,3-diones and catechols using air as an oxidant. The reactions can be carried out under mild reaction conditions without using toxic reagents. The products were obtained in yields ranging from 70 to 97% and with high purity. Byproducts were not formed. The structures of all products were unambiguously elucidated by NMR spectroscopic methods.
In the second part of this work laccase-initiated domino reactions between catechols and heterocyclic 1,3-dicarbonyls have been presented. Using pyridinones, quinolinones and thiocoumarin as substrates, the corresponding benzofuropyridinones, benzofuroquinolinones and thiocoumestans were being obtained. The reactions could be easily performed to deliver the products regioselctively in yields ranging between 55 and 98%. In contrast, polycyclic dispiropyrimidinones were exclusively formed when barbituric acid derivatives were employed as substrates. The unambiguous and complete structure elucidation of all products has been achieved by NMR spectroscopic methods (HSQMBC and band-selective HMBC) as well as by X-ray crystal structure analysis.
In the third part of this work laccase-catalyzed transformations between differently substituted hydroquinones and 1,3-dicarbonyls have been studied. These reactions provide a new and highly selective method for the formation of quinone bisadducts with two adjacent 1,3-dicarbonyl substituents. The only exception is the reaction of 2-chlorohydroquinone with 4-hydroxycoumarin which delivers a trisadduct. It is noteworthy that under different conditions the reaction between hydroquinones and 1,3-dicarbonyls resulted in the formation of benzofuran derivatives. The unambiguous structure elucidation of all products has been achieved by NMR spectroscopic methods including spin pattern analysis of the long-range coupled C=O carbons and 13C satellites analysis in 1H NMR spectra.
The domino reactions presented in this thesis allow for the efficient and selective synthesis of numerous heterocyclic systems as well as substituted p-benzoquinones under mild reaction conditions. In most cases the products can be isolated in good to very good yields and with high purity. For the structure elucidation of the products a wide range of NMR methods was used.

Die oxidative Phenolkupplung ist eine der grundlegenden Reaktionen der organischen Chemie. Im Gegensatz zu ihrer großen Bedeutung für die Biosynthese zahlreicher Naturstoffe spielt sie aufgrund häufig mangelnder Regio- und Stereoselektivität in der organischen Synthese bislang eher eine untergeordnete Rolle. Laccasen sind Oxidasen, mit denen sich unter Verwendung von O2 als Oxidationsmittel unter anderem auch oxidative Phenolkupplungen durchführen lassen. In dieser Arbeit wird an Hand von drei Beispielen belegt, dass sich Laccasen als effiziente Katalysatoren für regio- und stereoselektive Kupplungen von Phenolderivaten einsetzen lassen.
Im ersten Beispiel wurde die Laccase-katalysierte oxidative Dimerisierung von 2-Propenylsesamol zu Carpanon (a) untersucht. Die oxidative Cyclisierung beginnt mit einer Phenoloxidation, an die sich eine Radikalkupplung sowie eine intramolekulare Hetero-Diels-Alder-Reaktion anschließen. Experimente mit Laccasen und verschiedenen anderen Katalysatoren weisen darauf hin, dass die Diastereoselektivität der Carpanonbildung nicht vom Katalysator, sondern von der Doppelbindungsgeometrie des Substrats abhängt. Wurde die Reaktion mit (E)-2-Propenylsesamol als Substrat durchgeführt, isolierte man - unabhängig vom verwendeten Katalysator - neben dem Hauptprodukt Carpanon (a) immer auch ein zweites Produkt, und zwar das Carpanondiastereoisomer c. Die beiden Diastereoisomere wurden stets im Verhältnis 9:1 gebildet. Führte man die Reaktion dagegen mit (Z)-2-Propenylsesamol als Substrat durch, beobachtete man ein 5:1:4-Gemisch der drei Carpanondiastereoisomeren a, c und d.
Wurde die Oxidation von (E)-2-Propenylsesamol mit O2 in Abwesenheit eines Katalysators ausgeführt, kam es zur überraschenden Bildung der diastereoisomeren Benzopyrane a und b im Verhältnis von 3:2. Mechanistisch gesehen verläuft die Reaktion von (E)-2-Propenylsesamol hier als eine Domino Oxidation/intermolekulare Hetero-Diels-Alder-Reaktion.
Anschließend wurde die Laccase-katalysierte oxidative Kupplung von Sesamol, einem natürlich vorkommenden phenolischen Antioxidans, mit O2 als Oxidationsmittel studiert. Die Reaktion verlief mit guter Ausbeute unter Bildung eines bislang unbekannten Trimers als Hauptprodukt. Experimente mit verschiedenen Katalysatoren zeigten, dass der Verlauf der oxidativen Kupplung von Sesamol stark katalysatorabhängig ist.
Abschließend wurde die Phenolkupplung von di- und trisubstituierten Vanillidenderivaten mit Laccase als Katalysator und O2 als Oxidans untersucht. Die Dimerisierung von (E)-Ferulasäure verlief im Sinne einer 8,8?-Kupplung unter Bildung eines Dilactons. Bei der Reaktion der disubstituierten Vanillidenderivate kommt es im Rahmen einer 5,8?-Kupplung zur diastereoselektiven Bildung der Dihydrobenzo[b]furane in racemischer Form. In Gegensatz zu den disubstituierten Vanillidenderivaten isolierte man bei der Laccase-katalysierten Umsetzung der trisubstituierten Vanillidenderivate ausschließlich Biphenyle, die durch 5,5?-Kupplung entstehen.
The oxidative phenolic coupling is one of the fundamental reactions of organic chemistry. In contrast to its major role in the biosynthesis of numerous natural compounds the oxidative phenolic coupling is only of little importance in organic synthesis so far. This is due to its frequent lack of regio- and stereoselectivity. Laccases are oxidases which can be employed, amongst others, for the catalysis of oxidative phenolic couplings using O2 as the oxidant. This study highlights three examples which clearly demonstrate that laccases can be used as catalysts for regio- and stereoselective oxidative couplings of phenolic compounds.
The first example deals with the laccase-catalyzed oxidative dimerization of (E)-2-propenylsesamol to carpanone (a). The oxidative cyclization starts with a phenolic oxidation, which is followed by a radical coupling and an intramolecular hetero-Diels-Alder reaction. Experiments with laccases and a number of other catalysts indicate that the diastereoselectivity of the carpanone formation doesn´t depend on the nature of the catalyst but on the double bond geometry of the substrate. With (E)-2-propenylsesamol as the substrate, a 9:1-mixture of carpanone (a) and its diastereoisomer c was formed, irrespective of the catalyst used. When (Z)-2-propenylsesamol was used as the substrate, the formation of a 5:1:4-mixture of three diastereoisomers, i.e. a, c and d, was observed.
When the oxidation of (E)-2-propenylsesamol with O2 as the oxidant was run in the absence of any catalyst the diastereoisomeric benzopyrans a and b were obtained in a 3:2-ratio. From a mechanistic point of view, this reaction proceeds as a Domino oxidation/intermolecular hetero-Diels-Alder reaction.
The second example selected was the laccase-catalyzed oxidative coupling of sesamol, a naturally occurring phenolic antioxidant. Here, a so far unknown trimer was formed as the main product in good yield. Experiments with different catalysts indicated that the course of the oxidative coupling of sesamol depends strongly on the catalyst chosen.
Finally, the laccase-catalyzed phenolic coupling of di- and trisubstituted vanillidene derivatives with O2 as the oxidant was studied. The dimerization of (E)-ferulic acid proceeded as a 8,8?-coupling with formation of a dilactone. When the disubstituted vanillidene derivatives were reacted, the diastereoselective formation of the racemic dihydrobenzo[b]furans which can be understood as the products of a 5,8?-coupling mode were formed. In contrast to the disubstituted vanillidene derivatives, the laccase-catalyzed reaction of the trisubstituted vanillidene derivatives exclusively yielded biphenyls as the result of a 5,5?-coupling.

For the chemical analysis of food, drugs, and environmental samples it becomes more and more important to find substances of a certain (biological) activity. For this, several biological screening assays are available. One of the most versatile is the luminescent bacteria test according to an international norm (DIN EN ISO 11348), a rapid cuvette test on cytotoxicity. The assay employs the naturally bioluminescent bacterium Vibrio fischeri, which emits blue-green light under good living conditions. Because the energy-consuming luminescence metabolism is linked directly to the bacterium?s respiratory chain, a disturbance of the bacterium?s metabolism affects the luminescence, whereas the degree of toxicity is proportional to the luminescence inhibition.
Major advantage was achieved by coupling this biotest with previous separation by high-performance thin-layer chromatography (HPTLC), which allowed for a screening for individual components. The workflow consists of sample application onto an HPTLC plate, separation, drying the plate, application of the Vibrio fischeri suspension, and detection with a light-sensitive CCD camera. In the resulting image, dark zones on a brightly luminizing background indicate substances that affect the bacteria?s metabolism.
No suitable image evaluation program for the effective correction and quantitative evaluation of the image after Vibrio fischeri detection was available, which was regarded as a great disadvantage. In literature, adaptations of the special corrections based on the cuvette test calculations were described, including horizontal background correction and the recalculation of the sigmoid dose-response-relationship of the bacteria?s reaction. This served as a basis for the development of a new method using existing software which did not only perform the necessary calculations but was easy and convenient enough for use in routine evaluations.
Furthermore, the process of applying the aqueous bacteria suspension onto the HPTLC plates was improved. Usually, application was done by dipping with the help of a dipping device. Especially for polar substances, however, it was observed that substances can start to dissolve during this process, leading to blurring and tailing of the zones on the plate. A simple rolling device consisting of commercially available household articles was constructed. To compare rolling with dipping, octhilinone and methylparaben were chosen as test compounds. The results of rolling were far superior to dipping. However, manual rolling depended on the person who did it, and it was not possible to control pressure and velocity. To overcome this problem, a prototype of an automated rolling device was constructed and built.
After the successful process optimizations, the applicability of the HPTLC-bioluminescence assay was tested on commercial mouthwashes. Mouthwashes are likely to contain antimicrobial compounds, which are not necessarily indicated on the packaging. HPTLC with biodetection was used as a rapid screening method to detect zones of interest, which were further analyzed by conventional techniques like HPLC and GC. First, the reaction of Vibrio fischeri towards more than 40 standard substances was determined. This database was used for the analysis of six commercially available mouthwashes. It revealed that not only declared preservatives are used in mouthwashes, but also other antimicrobial compounds. These were especially constituents of essential oils having antibacterial properties (anethole, carvone, menthol, thymol), but are summarized as ?aroma?, which is in compliance with legal restrictions.
A most interesting question concerns the bacteria?s condition on the HPTLC plate. For the brightly luminizing background, it must be assumed that the bacteria are well alive. But no clear statement can be given for bacteria in the dark zones: they might be dead, inhibited (maybe only temporarily), or absent due to water repelling effects of the zone?s compound. A basic attempt to answer this question was made by applying a combination of classical microbiological techniques.
In this dissertation it could be shown that HPTLC coupled with Vibrio fischeri detection can successfully be used in practice and is well suited to complement conventional analytical techniques. This work is meant to serve as a guideline for further research and new applications.
Bei der chemischen Analyse von Lebensmitteln, Medikamenten und Umweltproben wird es immer wichtiger, Substanzen mit einer bestimmten (biologischen) Wirkung nachzuweisen. Hierfür stehen verschiedene biologische Screening-Assays zur Verfügung. Einer der universellsten ist der Leuchtbakterientest gemäß internationaler Norm (DIN EN ISO 11348), ein schneller Küvettentest auf Zytotoxizität. Für diesen Assay wird das von Natur aus biolumineszente Bakterium Vibrio fischeri eingesetzt, welches unter guten Lebensbedingungen blau-grünes Licht emittiert. Da der energieaufwändige Lumineszenz-Metabolismus direkt mit der Atmungskette des Bakteriums verknüpft ist, beeinflusst eine Störung des Bakterienstoffwechsels die Lumineszenz, wobei der Grad der Toxizität proportional zur Hemmung der Lumineszenz ist. Ein großer Fortschritt war die Kopplung dieses Biotests mit vorheriger Trennung durch die Hochleistungs-Dünnschichtchromatographie (HPTLC), da dies ein Screening auf Einzelsubstanzen ermöglicht. Der Arbeitsablauf besteht aus Probenauftragung auf eine HPTLC-Platte, Trennung, Trocknen der Platte, Aufbringen der Vibrio fischeri-Suspension und Detektion mit einer lichtempfindlichen CCD-Kamera. Im resultierenden Bild zeigen dunkle Zonen auf einem hell lumineszierenden Hintergrund Substanzen an, die den Bakterienstoffwechsel beeinflussen.
Für die effektive Korrektur und quantitative Auswertung des Bildes war kein geeignetes Bildauswerteprogramm vorhanden, was als großer Nachteil angesehen wurde. In der Literatur wurden Anpassungen der speziellen Korrekturen basierend auf den Berechnungen des Küvettentests beschrieben, einschließlich horizontaler Hintergrundkorrektur und Rückrechnung der sigmoiden Dosis-Wirkungsbeziehung der Bakterienreaktion. Dies diente als Basis für die Entwicklung einer neuen Methode unter Verwendung bestehender Software, die nicht nur die notwendigen Berechnungen durchführte, sondern auch einfach und komfortabel genug zu bedienen war, um sie routinemäßig für die Auswertung einzusetzen.
Weiterhin wurde das Aufbringen der wässrigen Bakteriensuspension auf die HPTLC-Platten verbessert. Üblicherweise wurde diese durch Tauchen mit Hilfe eines Tauchgerätes aufgebracht. Besonders bei polaren Substanzen wurde jedoch beobachtet, dass die Substanzen dabei angelöst werden können, was zu Verlaufen und Tailing der Zonen auf der Platte führte. Aus kommerziell erhältlichen Haushaltsartikeln wurde ein einfaches Walzgerät konstruiert. Um Walzen und Tauchen zu vergleichen, wurden Octhilinon und Methylparaben als Testsubstanzen gewählt. Die Ergebnisse mittels Walzen waren denen des Tauchens weit überlegen. Allerdings hing das manuelle Walzen von der ausführenden Person ab, und es war nicht möglich, Druck und Geschwindigkeit zu kontrollieren. Um dieses Problem zu bewältigen, wurde ein Prototyp eines automatischen Walzgerätes konstruiert und gebaut.
Nach diesen erfolgreichen Prozessoptimierungen wurde die Anwendbarkeit des HPTLC-Biolumineszenz-Assays an kommerziellen Mundspüllösungen getestet. Mundspüllösungen können antimikrobielle Bestandteile enthalten, welche nicht unbedingt auf der Packung angegeben sein müssen. HPTLC mit Biodetektion wurde als schnelle Screeningmethode eingesetzt, um die interessierenden Zonen zu detektieren, welche dann mit konventionellen Techniken wie HPLC und GC weiter untersucht wurden. Zunächst wurde die Reaktion von Vibrio fischeri auf mehr als 40 Standardsubstanzen bestimmt. Diese Datenbasis wurde für die Untersuchung von sechs kommerziell erhältlichen Mundspüllösungen verwendet. Es zeigte sich, dass in Mundspüllösungen nicht nur deklarierte Konservierungsstoffe, sondern auch andere antimikrobielle Substanzen verwendet werden. Diese waren hauptsächlich Bestandteile ätherischer Öle mit antibakteriellen Eigenschaften (Anethol, Carvon, Menthol, Thymol), die jedoch in Übereinstimmung mit den rechtlichen Vorschriften unter ?Aroma? zusammengefasst wurden.
Eine sehr interessante Frage betrifft das Schicksal der Bakterien auf der HPTLC-Platte. Für den hell leuchtenden Hintergrund muss angenommen werden, dass die Bakterien leben. Für die Bakterien in den dunklen Zonen kann jedoch keine klare Aussage getroffen werden: sie könnten tot, gehemmt (eventuell nur temporär), oder aufgrund wasserabstoßender Effekte der Substanz in der Zone nicht vorhanden sein. Durch Kombination klassischer mikrobiologischer Techniken wurde ein einfacher Versuch gemacht, diese Frage zu beantworten.
In dieser Dissertation konnte gezeigt werden, dass die Kopplung der HPTLC mit Vibrio fischeri-Detektion erfolgreich in der Praxis eingesetzt werden kann und die Methode zur Vervollständigung konventioneller Analysetechniken gut geeignet ist. Diese Arbeit soll als Leitfaden für die weitere Forschung und neue Anwendungen dienen.

The present thesis focuses on the influence of microwave irradiation and ionic liquids on the outcome of two well-known three component reactions, the Groebke reaction and the Povarov reaction. The first part of the thesis deals with the influence of microwaves and ionic liquids on the Groebke reaction. The reaction of 2-aminopyridines with aldehydes and isocyanides using montmorillonite as a catalyst in toluene under microwave conditions at 160°C delivers the corresponding imidazo[1,2-a]pyridines within only seven minutes with yields ranging from 16 to 98%. The organic solvent can be replaced by ionic liquids like imidazolium and guanidinium salts. With guanidinium salts, it is possible to perform the Groebke reaction in the absence of any other catalyst and solvent under microwave conditions. The second part of this work is about the extension of the scope of typical Groebke reactions by replacing the aldehyde component with a bifunctional 2-carboxybenzaldehyde. The reaction of 2-aminopyridines with isocyanides and 2-carboxybenzaldehydes with 20 mol% methanesulfonic acid as a catalyst in toluene under microwave conditions at 160°C affords the corresponding pyrido[2?,1?:2,3]imidazo[4,5-c]isoquinolin-5(6H)-ones with yields ranging between 35 and 68%. The new method can easily be performed, is robust, and highly efficient. The third part of the thesis is focused on the intermolecular Povarov reaction. Using the reaction between aniline, benzaldehyde, and 2,3-dihydrofuran as a model reaction, the influence of ionic liquids, such as imidazolium and guanidinium salts, and microwaves on the outcome of the Povarov reaction was evaluated. It was established that the model reaction can be promoted by imidazolium salts like [bmim]BF4 under thermal as well as under microwave conditions. The reaction temperature has a strong impact on the chemical yield and the diastereoselectivity of the model reaction. At lower temperatures the formation of the endo-isomer is favored. However, the influence of microwave irradiation on yield and selectivity is not very pronounced. The Povarov reaction can also be promoted by a great number of guanidinium salts. Reactions that were performed under thermal conditions in a sealed vial demonstrated that both the chemical yield and the diastereoselectivity of the reaction are strongly influenced by a) the structure of the guanidinium ion and the nature of the anion of the guanidinium salt, and b) the concentration of the guanidinium salt. Remarkably, the Povarov can also be performed successfully in the presence of only catalytic amounts of a guanidinium salt. Finally, it was demonstrated that the guanidinium salts can be recycled and reused several times without loss of reactivity.
Die vorliegende Dissertation beschäftigt sich vor allem mit dem Einfluss von Mikrowellenstrahlung und ionischen Flüssigkeiten auf zwei Dreikomponenten-Reaktionen, die Groebke- und die Povarov-Reaktion. Im ersten Teil der Dissertation wird der Einfluss von Mikrowellen und ionischen Flüssigkeiten auf die Groebke-Reaktion untersucht. Die Umsetzung von 2-Aminopyridinen mit Aldehyden und Isocyaniden mit Montmorillonit als Katalysator in Toluol unter Mikrowellenbedingungen bei 160°C liefert die entsprechenden Imidazo[1,2-a]pyridine innerhalb von nur sieben Minuten mit Ausbeuten zwischen 16 und 98%. Das organische Lösungsmittel kann durch ionische Flüssigkeiten wie Imidazolium- und Guanidiniumsalze ersetzt werden. Mit Guanidiniumsalzen ist es möglich, die Groebke-Reaktion in Abwesenheit irgendeines zusätzlichen Katalysators oder Lösungsmittels unter Mikrowellenbedingungen durchzuführen. Der zweite Teil dieser Arbeit befasst sich mit der Erweiterung des Anwendungsbereichs der klassischen Groebke-Reaktion durch Ersatz der Aldehydkomponente durch einen bifunktionalen 2-Carboxybenzaldehyd. Die Reaktion von 2-Aminopyridinen mit Isocyaniden und 2-Carboxybenzaldehyden in Gegenwart von 20 mol% Methansulfonsäure als Katalysator in Toluol unter Mikrowellenbedingungen bei 160°C ergibt die entsprechenden Pyrido[2?,1?:2,3]imidazo[4,5-c]isochinolin-5(6H)-one mit Ausbeuten zwischen 35 und 68%. Die neue Methode kann leicht durchgeführt werden, ist robust und hoch effizient. Der dritte Teil dieser Dissertation beschäftigt sich mit der intermolekularen Povarov-Reaktion. Am Beispiel der Umsetzung zwischen Anilin, Benzaldehyd und 2,3-Dihydrofuran als Modellreaktion wurde der Einfluss von ionischen Flüssigkeiten wie Imidazolium- und Guanidiniumsalzen sowie von Mikrowellen auf den Verlauf der Povarov-Reaktion untersucht. Man fand, dass die Modellreaktion sowohl unter thermischen als auch unter Mikrowellenbedingungen durch Imidazoliumsalze wie [bmim]BF4 katalysiert werden kann. Die Reaktionstemperatur übt einen starken Einfluss auf die chemische Ausbeute und die Diastereoselektivität der Modellreaktion aus. Bei niedrigen Temperaturen wird die Bildung des endo-Isomers bevorzugt. Dagegen ist der Einfluss der Mikrowellenstrahlung auf Ausbeute und Selektivität nicht sehr ausgeprägt. Die Povarov-Reaktion kann auch durch viele Guanidiniumsalze initiiert werden. Am Beispiel der Umsetzungen unter thermischen Bedingungen in einem verschlossenen Röhrchen ließ sich zeigen, dass sowohl a) die Struktur des Guanidiniumions und die Art des Anions des Guanidiniumsalzes als auch b) die Konzentration des Guanidiniumsalzes einen starken Einfluss auf die chemische Ausbeute und die Diastereoselektivität ausüben. Bemerkenswert ist, dass sich die Povarov-Reaktion auch in Gegenwart von nur katalytischen Mengen eines Guanidiniumsalzes erfolgreich realisieren lässt. Abschließend wurde gezeigt, dass die Guanidiniumsalze recycelt und mehrere Male ohne Verlust der Reaktivität wiederverwendet werden können.

The onset of life on Earth was preceded by an abiotic chemistry in which complex molecules were formed from simpler ones. In the presence of energy sources such as UV radiation, lightning and geothermal energy, a wide range of organic compounds probably formed on the young Earth. Stanley Miller was the first to study this scenario experimentally. He showed that amino acids were synthesized under simulated conditions of the primitive Earth?s atmosphere. Initially, it was believed that the Earth?s early atmosphere contained high concentrations of CH4, NH3, CO, and H2 and was thus strongly reducing. However, later it was assumed that the early atmosphere was redox neutral and was composed of N2, CO2, and H2O as main constituents. Isotopic data from zircons indicate that liquid water might have been present already around 4.2 billion years ago. So called banded iron formations confirm the presence of liquid water at least 3.8 billion years ago. The early geological histories of Earth and Mars were probably very similar. About 4 billion years ago, both planets had liquid water, volcanoes, and a dense atmosphere without free oxygen, and they experienced intense meteoritic and cometary impacts. Therefore, the simulation experiments described in the present thesis may also be relevant to the early Mars. Among the possible prebiotic molecules, amino acids are generally considered especially important for the origin of life. The main reason for this is that they serve as building blocks of proteins which are the pillars of metabolism in all organisms. There is practically no doubt that amino acids were present on the young Earth. They originated from endogenous and exogenous (i.e. extraterrestrial) sources. Glycine is the most abundant amino acid in carbonaceous meteorites and Miller-type experiments.
In the present work, the deep black residue was studied that forms when glycine is heated at 200 °C. Similar residues have been named ?thermo-melanoid? by others. The experiments were performed under a pure nitrogen atmosphere in order to simulate the oxygen-free early atmosphere of the Earth. It was found that the formation of the thermomelanoid from neat glycine started at 160 °C and was relatively fast and complete at 200 °C. However, the residues that formed at high temperatures (250?350 °C) were different from the thermo-melanoid. The thermo-melanoid was also present in the residues obtained by heating the glycine homopeptides 2,5-diketopiperazine (DKP), diglycine, triglycine, and tetraglycine at 200 °C. In contrast, penta- and hexaglycine remained almost unreacted at this temperature. Deuterolysis experiments revealed that C=C bonds are a characteristic structural feature of the thermo-melanoid. These bonds form by an unusual condensation reaction between C=O and CH2 groups. Glycine, DKP, and diglycine were released during hydrolysis of the thermomelanoid in water at 100 °C. In these experiments, the thermo-melanoid slowly dissolved. After 10 days, for example, a mass loss of ~75 % was observed. Therefore, the thermomelanoid can be regarded as a kind of storage form of glycine and glycine oligopeptides. The lower solubility of the thermo-melanoid as compared to glycine and its homopeptides may have influenced the distribution of glycine units on the early Earth. Moreover, additional experiments have shown that the thermo-melanoid mixed with soil continuously produced a higher amount of CO2 during a six-months period than samples without the thermo-melanoid. Obviously, the thermo-melanoid was decomposed in the soil. The decomposition was probably caused by microorganisms. Therefore, one can hypothesize that the thermomelanoid could have served as nutrient for early heterotrophic (pre-)organisms.
The salt concentration of the late Hadean/early Archean ocean was at least twice as high as the concentration in the present-day oceans. There are good reasons to assume that the ions were Na+, K+, Ca2+, Mg2+, and Cl?. SO4 2? and PO4 3? were possibly not present in significant concentrations as the early atmosphere of the Earth was anoxic. In relation to this, the thermal behavior of glycine was investigated in the presence of various salts. It was found that glycine changed from the initial α- to the γ-modification when it crystallized together with NaCl and NaCl?KCl mixtures. At 200 °C, the glycine that was embedded in the NaCl or NaCl?KCl salt crusts transformed into the thermo-melanoid and a small amount of DKP. Only ~5 % of unreacted glycine was left after seven days in the presence of NaCl. The results showed that the presence of these salts and the change in the modification were nearly ineffective in protecting glycine from transformation into the thermo-melanoid. In contrast to NaCl and KCl, CaCl2 formed a coordination compound with glycine, namely CaCl2(Hgly) ⋅ H2O, when solutions of CaCl2 ⋅ 2H2O and glycine were evaporated. It was found that more than 90 % of the glycine were still present in CaCl2(Hgly) ⋅ H2O after heating at 200 °C for seven days. The coordination of glycine to Ca2+ prevented the transformation of glycine into the thermo-melanoid up to 250 °C. Yusenko et al. reported that at 350 °C, small volatile N-heterocycles such as pyrroles formed from CaCl2(Hgly) ⋅ H2O. Pyrroles are the building blocks of porphyrin-type biomolecules such as cytochromes and chlorophylls. CaCl2(Hgly) ⋅ H2O was also identified in mixtures of glycine with artificial sea salt (AS) prepared from NaCl (705 mmol), KCl (15 mmol), MgCl2 ⋅ 6H2O (80 mmol), CaCl2 ⋅ 2H2O (15 mmol), and glycine (10 mmol). About 84 % of the initial glycine had survived after heating an AS?Hgly mixture for seven days at 200 °C. In contrast, neither complex formation nor change in the modification of glycine was observed in gypsum?Hgly and MgCO3?Hgly mixtures.
Clay minerals are mainly produced by the weathering of volcanic rock. They are not only found on Earth, but also on Mars. A possible role of clay minerals in chemical evolution was first suggested by Bernal more than half a century ago. In the present work, the thermal behavior of glycine embedded in smectites (Ca-montmorillonite, Na-montmorillonite, and nontronite) and kaolinite was investigated. The glycine-loaded clay minerals were heated at 200 and 250 °C for two days. HPLC and MALDI?TOF/TOF MS analyses of glycine-loaded Ca-montmorillonite that had been heated at 200 °C showed the presence of unreacted glycine, DKP, and linear peptides up to decaglycine. The comparison between the smectite clay minerals revealed that glycine was best protected by Ca-montmorillonite. ~63 % of the amino acid survived in its free form at 200 °C. This was followed by Na-montmorillonite (~53 %) and nontronite (~39 %) under similar experimental conditions. These results demonstrated that smectite clay minerals protect glycine from complete decomposition and sublimation, partly by promoting its polymerization. In contrast to smectites, kaolinite has no interlayer spaces available for intercalation. Therefore, glycine is only attached to the surface of the kaolinite particles. Sublimation of glycine and newly formed DKP was observed when kaolinite mixed with glycine was heated at 200 and 250 °C. All investigated clay minerals prevented the transformation of glycine into the thermo-melanoid during thermal treatments.
Various heating experiments were conducted with mixtures of glycine and volcanic rock (basaltic sand from the island of La Réunion, Indian Ocean) or Martian soil simulants (JSC Mars-1A, P-MRS, and S-MRS). Glycine and DKP were identified in the residues and sublimates after heating basaltic sand?Hgly and JSC Mars-1A?Hgly at 200 °C for two days. Additionally, the thermo-melanoid was found in the residue of basaltic sand?Hgly. JSC Mars-1A contains mainly volcanic glass. Forsterite (Mg2SiO4) was identified as the major crystalline mineral in the basaltic sand. Glycine cannot be intercalated in JSC Mars-1A and basaltic sand as they have no clay minerals. As a result, glycine in these two matrices undergoes thermal alterations similar to neat glycine. In contrast, glycine, DKP, and linear peptides from di- to hexaglycine were detected after heating a P-MRS?glycine mixture. This observation can be easily explained by the fact that P-MRS contains 70 % of clay minerals that protect the amino acid from complete decomposition and thus allow the formation of larger peptides. The S-MRS?glycine residue contained only DKP, glycine, and diglycine, obviously because the mineral matrix consisted only of rock, anhydrous iron oxides, and gypsum, but not clay minerals. These experiments again demonstrated the influence of clay minerals on the behavior of glycine when exposed to higher temperatures.
Another focus of the work was on the thermal behavior of chiral amino acids intercalated in Ca-montmorillonite. The heating experiments were conducted with different L-enantiomeric excesses (ee) of alanine [L-ee = 0 (i.e. racemic), 4, 20, 50, and 100 %] under a pure nitrogen atmosphere. The residues were analyzed by GC-MS/FID after derivatization. It was found that the racemization process was fast during the first 2?3 days and thereafter slowed down considerably. After eight weeks at 200 °C, the residues still contained 17.5?25.0 % of the respective starting L-ee. Complete racemization of L-alanine was not observed even after 24 weeks of heating. It was also found that, as expected, Camontmorillonite did not have any specific preference for the formation of either the D- or L-enantiomer. Interestingly, it could be observed that L-isovaline influenced the racemization of alanine. The presence of L-isovaline increased the rate of formation of D-alanine. In addition, higher temperatures greatly accelerated the racemization. For instance, after eight weeks at 220 °C, 85 % of the initial L-ee of alanine had been lost by racemization, whereas at 120 °C only 25 % racemization was observed. These experiments made use of the fact that Ca-montmorillonite largely protects amino acids from sublimation. In contrast, neat amino acids such as alanine undergo considerable sublimation in a few hours or less, depending on the temperature. Using a racemization kinetics model, it was estimated that L-alanine can survive in Ca-montmorillonite at elevated temperatures for years.
In the literature, there are several reports on enantiomeric excesses of certain amino acids in meteorites. In relation to this, experiments were performed to demonstrate the enantiomeric enrichment of amino acids by partial sublimation. After 3 and 24 hours at 200 °C, the L-ee of alanine and valine increased in the sublimation residue, whereas the L-ee of the sublimates was lower than the initial one. Thus, it seems that racemic alanine and racemic valine crystals are more volatile than enantiomerically pure crystals. It may be assumed that similar processes take place during the atmospheric entry of meteorites and in the aqueous alteration phase of asteroids.
The experimental results described in the present thesis suggest that various modes of interaction of amino acids with inorganic matrices such as salt mixtures and clay minerals existed on the young Earth. These results may help to better understand some of the processes of the prebiotic chemical evolution.
Dem Beginn des Lebens auf der Erde ging eine abiotische Chemie voraus, in der komplexe Moleküle aus einfacheren gebildet wurden. In Gegenwart von Energiequellen wie UVStrahlung, Blitzen und geothermaler Energie bildete sich wahrscheinlich eine breite Palette organischer Verbindungen auf der jungen Erde. Stanley Miller war der erste, der dieses Szenario experimentell erforschte. Er hat gezeigt, dass sich unter Bedingungen, die die frühe Erdatmosphäre simulierten, Aminosäuren bildeten. Anfänglich wurde angenommen, dass die frühe Atmosphäre der Erde hohe Konzentrationen an CH4, NH3, CO und H2 enthielt und daher stark reduzierend war. Später ging man jedoch davon aus, dass die frühe Atmosphäre redoxneutral und aus den Hauptbestandteilen N2, CO2 und H2O zusammengesetzt war. Isotopenmesswerte aus Zirkonen zeigen, dass flüssiges Wasser schon vor etwa 4,2 Milliarden Jahren vorhanden gewesen sein könnte. So genannte ?Banded Iron Formations? bestätigen die Anwesenheit von flüssigem Wasser seit mindestens 3,8 Milliarden Jahren. Die frühe geologische Entwicklung der Erde und des Mars verlief wahrscheinlich sehr ähnlich. Vor ungefähr 4 Milliarden Jahren besaßen beide Planeten flüssiges Wasser, Vulkane und eine dichte Atmosphäre ohne freien Sauerstoff und erlebten zahlreiche Einschläge von Meteoriten und Kometen. Daher dürften die Simulationsexperimente, die in der vorliegenden Arbeit beschrieben werden, auch für den frühen Mars relevant sein. Unter den möglichen präbiotischen Molekülen werden Aminosäuren allgemein als besonders wichtig für den Ursprung des Lebens angesehen. Dies wird hauptsächlich deshalb angenommen, weil sie als Bausteine für Proteine dienen, welche in allen Organismen die Grundlage des Stoffwechsels darstellen. Es gibt praktisch keinen Zweifel daran, dass Aminosäuren auf der jungen Erde vorhanden waren. Sie stammten aus endogenen und exogenen (d. h. außerirdischen) Quellen. Glycin ist die häufigste Aminosäure in kohligen Meteoriten und in Experimenten des Miller-Typs.
In der vorliegenden Arbeit wurde der tiefschwarze Rückstand untersucht, der sich bildet, wenn Glycin auf 200 °C erhitzt wird. Ähnliche Rückstände sind von anderen als ?Thermomelanoid? bezeichnet worden. Die Versuche wurden unter reiner Stickstoffatmosphäre durchgeführt, um die sauerstofffreie frühe Atmosphäre der Erde zu simulieren. Es wurde festgestellt, dass die Bildung des Thermomelanoids aus reinem Glycin bei 160 °C beginnt und bei 200 °C verhältnismäßig schnell und vollständig ist. Die Rückstände, die sich bei hohen Temperaturen (250?350 °C) bildeten, unterschieden sich jedoch vom Thermomelanoid. Das Thermomelanoid war auch in den Rückständen vorhanden, die durch Erhitzen 200 °C erhalten wurden. Im Gegensatz dazu zeigten Penta- und Hexaglycin bei dieser Temperatur fast keine Reaktion. Deuterolyseversuche haben gezeigt, dass C=CDoppelbindungen ein charakteristisches strukturelles Merkmal des Thermomelanoids sind. Diese Bindungen bilden sich durch eine ungewöhnliche Kondensationsreaktion zwischen C=O- und CH2-Gruppen. Während der Hydrolyse des Thermomelanoids bei 100 °C in Wasser wurden Glycin, DKP und Diglycin freigesetzt. In diesen Versuchen löste sich das Thermomelanoid allmählich auf. Nach 10 Tagen wurde beispielsweise ein Masseverlust von etwa 75 % beobachtet. Daher kann das Thermomelanoid als eine Art Speicherform von Glycin und Glycin-Oligopeptiden betrachtet werden. Die geringere Löslichkeit des Thermomelanoids im Vergleich zum Glycin und seinen Homopeptiden könnte die Verteilung von Glycin-Einheiten auf der frühen Erde beeinflusst haben. Außerdem haben weitere Versuche gezeigt, dass das Thermomelanoid mit Boden vermischt in einem Zeitraum von sechs Monaten kontinuierlich eine größere Menge CO2 freisetzte als Vergleichsproben ohne das Thermomelanoid. Offensichtlich wurde das Thermomelanoid in der Erde zersetzt. Die Zersetzung wurde wahrscheinlich von Mikroorganismen verursacht. Daher kann man vermuten, dass das Thermomelanoid frühen heterotrophen (Vor-)Organismen als Nährstoff gedient haben könnte.
Die Salzkonzentration des Ozeans im späten Hadaikum/frühen Archaikum war mindestens doppelt so hoch wie die Konzentration in den heutigen Ozeanen. Es gibt gute Gründe anzunehmen, dass es sich bei den Ionen um Na+, K+, Ca2+, Mg2+ und Cl? handelte. SO4 2? und PO4 3? waren möglicherweise nicht in wesentlichen Konzentrationen vorhanden, da die frühe Atmosphäre der Erde nichtoxidierend war. In Zusammenhang damit wurde das thermische Verhalten von Glycin in Anwesenheit verschiedener Salze untersucht. Es konnte festgestellt werden, dass sich Glycin von der ursprünglichen α-Modifikation in die γ-Modifikation umwandelte, wenn es gemeinsam mit NaCl oder NaCl?KCl Mischungen auskristallisierte. Bei 200 °C wandelte sich das Glycin, das in den NaCl- oder den NaCl? KCl-Salzkrusten eingebettet war, in das Thermomelanoid und wenig DKP um. In Anwesenheit von NaCl verblieben nach sieben Tagen nur etwa 5 % unreagiertes Glycin. Die Ergebnisse haben gezeigt, dass die Anwesenheit dieser Salze und die Modifikationsänderung so gut wie nicht dazu in der Lage waren, Glycin vor der Umwandlung in das Thermomelanoid zu schützen. Im Gegensatz zu NaCl und KCl bildete CaCl2 eine Koordinationsverbindung mit Glycin, nämlich CaCl2(Hgly) ⋅ H2O, wenn man Lösungen von CaCl2 ⋅ 2H2O und Glycin eintrocknen ließ. Es wurde festgestellt, dass mehr als 90 % des Glycins im CaCl2(Hgly) ⋅ H2O noch vorhanden waren, nachdem man es für sieben Tage bei 200 °C erhitzt hatte. Die Koordination des Glycins an Ca2+ verhinderte die Umwandlung der Aminosäure in das Thermomelanoid bis hinauf zu 250 °C. Yusenko et al. berichteten, dass sich aus CaCl2(Hgly) ⋅ H2O bei 350 °C kleine flüchtige N-Heterozyklen wie zum Beispiel Pyrrole bilden. Pyrrole sind die Bausteine von Biomolekülen des Porphyrin-Typs, wie zum Beispiel Cytochrome und Chlorophylle. CaCl2(Hgly) ⋅ H2O wurde auch in Mischungen von Glycin mit künstlichem Meersalz (AS) nachgewiesen, die aus NaCl (705 mmol), KCl (15 mmol), MgCl2 ⋅ 6H2O (80 mmol), CaCl2 ⋅ 2H2O (15 mmol) und Glycin (10 mmol) hergestellt wurden. Ungefähr 84 % des anfänglichen Glycins war noch vorhanden, nachdem eine AS?Hgly-Mischung für sieben Tage bei 200 °C erhitzt worden war. Im Gegensatz dazu konnten in Gips?Hgly- und MgCO3?Hgly-Mischungen weder Komplexbildung noch eine Modifikationsänderung des Glycins beobachtet werden.
Tonminerale entstehen hauptsächlich beim Verwittern vulkanischen Gesteins. Man findet sie nicht nur auf der Erde, sondern auch auf dem Mars. Vor mehr als einem halben Jahrhundert wies erstmals Bernal auf eine mögliche Rolle der Tonminerale in der chemischen Evolution hin. In der vorliegenden Arbeit wurde das thermische Verhalten von Glycin nach der Einbettung in Smektite (Ca-Montmorillonit, Na-Montmorillonit und Nontronit) und Kaolinit untersucht. Die glycinbeladenen Tonminerale wurden für zwei Tage bei 200 und 250 °C erhitzt. HPLC- und MALDI-TOF/TOF-MS-Analysen von glycinbeladenem Ca- Montmorillonit, das bei 200 °C erhitzt worden war, zeigten das Vorhandensein von nicht reagiertem Glycin, DKP und linearen Peptiden bis zum Decaglycin. Der Vergleich zwischen den Smektit-Tonmineralen ließ erkennen, dass Glycin am besten durch Ca-Montmorillonit geschützt wurde. Bei 200 °C waren 63 % der Aminosäure in unveränderter Form erhalten geblieben. Dem folgten Na-Montmorillonit (~53 %) und Nontronit (~39 %) unter ähnlichen experimentellen Bedingungen. Diese Ergebnisse haben gezeigt, dass Smektit-Tonminerale das Glycin vor vollständiger Zersetzung und Sublimation schützen, teilweise indem sie seine Polymerisation begünstigen. Im Gegensatz zu den Smektiten hat Kaolinit keine Zwischenschichträume, in denen Intercalation stattfinden kann. Daher wird Glycin nur an die Oberfläche der Kaolinitpartikel angelagert. Als Kaolinit, das mit Glycin vermischt worden war, auf 200 und 250 °C erhitzt wurde, wurde die Sublimation von Glycin und neu gebildetem DKP beobachtet. Alle untersuchten Tonminerale verhinderten, dass sich das Glycin während der thermischen Behandlungen in das Thermomelanoid unwandelte.
In verschiedenen Experimenten wurden Mischungen aus Glycin und Vulkangestein (Basaltsand von der Insel La Réunion, Indischer Ozean) oder simuliertem Marsboden (JSC Mars-1A, P-MRS und S-MRS) erhitzt. In den Rückständen und Sublimaten wurden Glycin und DKP identifiziert, nachdem Basaltsand?Hgly und JSC Mars-1A?Hgly für zwei Tage bei 200 °C erhitzt worden waren. Zusätzlich wurde das Thermomelanoid im Rückstand des Basaltsandes mit Hgly gefunden. JSC Mars-1A enthält hauptsächlich vulkanisches Glas. Forsterit (Mg2SiO4) wurde als das hauptsächliche kristalline Mineral im Basaltsand identifiziert. Glycin kann in JSC Mars-1A und im Basaltsand nicht intercaliert werden, da sie keine Tonminerale enthalten. Demzufolge erfährt Glycin in diesen zwei Matrizes ähnliche thermische Veränderungen wie das reine Glycin. Im Gegensatz dazu wurden nach dem Erhitzen einer P-MRS?Glycin-Mischung Glycin, DKP und lineare Peptide vom Di- bis zum Hexaglycin gefunden. Diese Beobachtung kann leicht damit erklärt werden, dass P-MRS 70 % Tonminerale enthält, die die Aminosäure vor vollständiger Zersetzung schützen und folglich die Bildung der größerer Peptide erlauben. Der S-MRS?Glycin-Rückstand enthielt nur DKP, Glycin und Diglycin, offensichtlich weil die mineralische Matrix nur aus Gesteinen, wasserfreien Eisenoxiden und Gips bestand, aber nicht aus Tonmineralen. Diese Versuche demonstrierten erneut den Einfluss von Tonmineralen auf das Verhalten von Glycin, wenn es höheren Temperaturen ausgesetzt ist.
Einen weiteren Schwerpunkt der Arbeit bildete das thermische Verhalten chiraler Aminosäuren, die in Ca-Montmorillonit intercaliert waren. Die Experimente wurden mit verschiedenen L-Enantiomerenüberschüssen (ee) von Alanin [L-ee = 0 (d. h. racemisch), 4, 20, 50 und 100 %] in einer reinen Stickstoffatmosphäre durchgeführt. Nach Derivatisierung wurden die Rückstände mittels GC-MS/FID analysiert. Es wurde festgestellt, dass der Racemisierungsvorgang während der ersten 2?3 Tage schnell verlief und danach beträchtlich langsamer wurde. Nach acht Wochen bei 200 °C enthielten die Rückstände noch 17.5?25.0 % des jeweiligen Start-L-ee. Eine vollständige Racemisierung des L-Alanins konnte selbst nach 24 Wochen Erhitzen nicht beobachtet werden. Es wurde außerdem festgestellt, dass Ca- Montmorillonit, wie erwartet, weder für die Bildung des D- noch des L-Enantiomers eine spezifische Präferenz aufwies. Interessanterweise konnte beobachtet werden, dass L-Isovalin die Racemisierung des Alanins beeinflusste. Die Anwesenheit von L-Isovalin erhöhte die Bildungsgeschwindigkeit von D-Alanin. Des Weiteren beschleunigten höhere Temperaturen die Racemisierung sehr stark. Zum Beispiel waren nach acht Wochen bei 220 °C 85 % des ursprünglichen L-ee durch Racemisierung verloren gegangen, während bei 120 °C nur 25 % Racemisierung beobachtet wurde. Diese Versuche nutzten die Tatsache, dass Ca-Montmorillonit Aminosäuren weitgehend vor Sublimation schützt. Im Gegensatz dazu erfahren reine Aminosäuren wie zum Beispiel Alanin beträchtliche, temperaturabhängige Sublimation innerhalb einiger Stunden oder weniger. Mithilfe eines Modells für die Kinetik der Racemisierung ließ sich abschätzen, dass L-Alanin in Ca-Montmorillonit bei erhöhten Temperaturen jahrelang überdauern kann.
In der Literatur gibt es zahlreiche Berichte über Enantiomerenüberschüsse bestimmter Aminosäuren in Meteoriten. In Zusammenhang damit wurden Versuche durchgeführt, um die Enantiomerenanreicherung von Aminosäuren durch partielle Sublimation zu zeigen. Nach 3 und 24 Stunden bei 200 °C hatte sich der L-ee von Alanin und Valin im Sublimationsrückstand erhöht, während der L-ee im Sublimat geringer war als der anfängliche. Folglich scheint es, dass Kristalle des racemischen Alanins und racemischen Valins flüchtiger sind als enantiomerenreine Kristalle. Man kann annehmen, dass ähnliche Prozesse während des Eintritts von Meteoriten in die Atmosphäre und in der ?Aqueous
Alteration?-Phase von Asteroiden stattfinden.
Die experimentellen Ergebnisse, die in der vorliegenden Arbeit beschrieben werden, legen nahe, dass auf der jungen Erde vielfältige Möglichkeiten der Wechselwirkung von Aminosäuren mit anorganischen Matrizes, wie zum Beispiel Salzmischungen und Tonmineralen, existierten. Diese Ergebnisse dürften dazu beitragen, einige der Vorgänge der präbiotischen chemischen Evolution besser zu verstehen.

Currently, the most serious problems in pesticide residue analysis by liquid chromatography (LC) or gas chromatography (GC) coupled to mass spectrometry (MS) concern the so-called matrix effects. The most common way to avoid these effects is the application of matrix-matched calibration standards. Nevertheless, an efficient clean-up undoubtedly is the best way to prevent matrix effects in multi-residue analysis of pesticides in food by LCMS or GCMS.
For a totally new powerful clean-up method, called high-throughput planar solid phase extraction (HTpSPE), highly automated planar chromatographic tools were applied to remove co-extracted matrix substances entirely and to eliminate any kind of matrix related effects. For sample extraction, the quick, easy, cheap, effective, rugged, and safe (QuEChERS) method was used to initially collect pesticides from fruits and vegetables. The received acetonitrile extracts were applied directly for the development of the novel HTpSPE clean-up. Thin-layer chromatography (TLC) was used to completely separate pesticides from matrix compounds and to focus them into a sharp zone. A two-fold development on amino-modified silica gel thin-layers with acetonitrile for the first development, and acetone for the second development in the backwards direction, was evaluated to perform the best clean-up result and collect the pesticides in a sharp, single target zone. To easily locate the pesticide zone, the Sudan II dye was added to the extracts. Following this clean-up, the target zones (pesticides) were eluted by the TLCMS interface into vials for the LCMS determination. HTpSPE resulted in extracts which were nearly free of co-extracted matrix and matrix effects, as shown for seven chemically representative pesticides (acetamiprid, azoxystrobin, chlorpyrifos, fenarimol, mepanipyrim, penconazole, and pirimicarb) in four different fruit and vegetable matrices (apples, cucumbers, red grapes, and tomatoes). Thanks to the very clean HTpSPE extracts, calibration can simply be performed with pure solvent standards and the quantitation by LCMS provided excellent mean recoveries and relative standard deviations.
In addition, tea samples as rather challenging matrices were chosen to apply for HTpSPE. The matrix load of tea extracts generally was too high for the available thin-layer capacity and the selectivity of the amino-modified phase was not suitable for the separation of caffeine and further matrix compounds from the target analytes (pesticides). By modifying the sample extraction, adding a pre-cleaning by dispersive solid phase extraction (dSPE) and changing the thin-layer phase to normal phase silica gel, the complete separation of pesticides and tea matrix components was possible, when again a two-fold development was applied. Caffeine and other alkaloids were completely removed. The effectiveness of HTpSPE was demonstrated by LCMS/MS calibration curves from matrix-matched and solvent standards, which were nearly identical and by very good mean recoveries, calculated against pure solvent standards. Concerning all validation parameters, the new acetonitrile-HTpSPE procedure for tea samples was superior to the QuEChERS-dSPE method and offered highly successful results.
In recent years, large-scale screening in pesticide residue analysis has gained more and more importance. Keeping this in mind, a screening strategy for HTpSPE extracts, using a high-resolution MS, was developed to analyze the cleaned extracts directly for pesticide residues without a liquid chromatographic separation. By this hyphenation, a completely new microliter-flow injection analysistime-of-flight mass spectrometry (µL-FIATOFMS) screening was introduced. The novel HTpSPEµL-FIATOFMS approach enabled the detection of all pesticides simultaneously in a single mass spectrum within a few minutes. The obtained mass spectra were nearly free of matrix compounds, which is especially the great benefit of the effective HTpSPE clean-up. Recovery studies by HTpSPEµL-FIATOFMS against solvent standards for the matrices and pesticides under study provided excellent results, using the mass signal intensities under the entire FIA sample peak. HTpSPE clearly showed superior results concerning every tested parameter than dSPE. With the help of a self-constructed mass database searching tool, all spiked pesticides were detected and correctly identified, while only very low numbers of false-positive findings occurred. Furthermore, a non-target screening approach was successfully implemented by slightly changing the database searching process, offering a mass list of all substances, which are present in the injected extracts but not included in the mass database. Finally, the new HTpSPEµL-FIATOFMS screening was successfully applied to several real samples, when the identified pesticides were quite identical compared to results of LCMS/MS analysis of the QuEChERS-dSPE extracts.
In der Rückstandsanalytik von Pestiziden bereiten Matrixeffekte bei der flüssigkeits- oder gaschromatographischen Bestimmung mittels Massenspektrometrie (LCMS oder GCMS) die größten Probleme. Standardmäßig versucht man, diese durch Verwendung von matrix-matched Kalibrierstandards zu kompensieren. Trotzdem ist eine effiziente Reinigung der Extrakte zweifelsohne der beste Weg, um diese Effekte in der Pestizidrückstandsanalytik in Lebensmitteln mittels LCMS oder GCMS zu vermeiden.
Für die Einführung einer völlig neuen, leistungsfähigen Clean-up Methode, der so genannten high-throughput planar solid phase extraction (HTpSPE), wurden hoch automatisierte planar-chromatographische Werkzeuge eingesetzt, um die mitextrahierte Matrix zu entfernen und matrixbedingten Effekten zu beseitigen. Die Probenextraktion erfolgte gemäß der QuEChERS-Methode. Die Acetonitril-Extrakte wurden direkt für die Entwicklung der neuen HTpSPE Reinigung verwendet. Dieses nutzt die Dünnschicht-Chromatographie (DC), um Pestizide und Matrix vollständig voneinander zu trennen und die Wirkstoffe in einer scharfen Zone zu sammeln. Das beste Clean-up Ergebnis ergab eine 2-fache Entwicklung auf Amino-modifizierten DC-Schichten. Die erste Entwicklung erfolgte mit Acetonitril, die zweite Entwicklung mit Aceton in der Rückwärtsrichtung. Vor der HTpSPE wurde den Extrakten der Farbstoff Sudan II als visueller Marker hinzugefügt, wodurch die Pestizidzone einfach zu erkennen ist. Nach diesem Clean-up wurden die Zielzonen mittels des TLCMS Interface in Gläschen eluiert und mittels LCMS analysiert. Nach dem HTpSPE Clean-up waren die Extrakte nahezu frei von mitextrahierter Matrix und Matrixeffekte. Dies wurde erfolgreich anhand von sieben chemisch repräsentativen Pestiziden (Acetamiprid, Azoxystrobin, Chlorpyrifos, Fenarimol, Mepanipyrim, Penconazol und Pirimicarb) in vier Obst- und Gemüsematrices (Äpfel, Gurken, roten Trauben und Tomaten) gezeigt. Dank der sehr sauberen HTpSPE-Extrakte kann die Quantifizierung einfach mit reinen Lösungsmittelstandards erfolgen. LCMS Messungen lieferten exzellente mittlere Wiederfindungen und relative Standardabweichungen.
Für eine weitere Anwendung wurden Teeproben als äußerst anspruchsvolle Matrices ausgewählt. Die Matrixfracht der Extrakte war für die Kapazität der DC-Schichten zu hoch und die Selektivität der Aminophase war nicht geeignet, um Koffein und weitere Matrixverbindungen von den Zielsubstanzen (Pestiziden) abzutrennen. Durch die Modifizierung der Extraktion, einer Vorreinigung mittels dispersiver Festphasenextraktion (dSPE) und dem Wechsel der DC-Phase zu unmodifiziertem Kieselgel konnte die vollständige Abtrennung der Matrix von den Pestiziden erreicht werden, wieder in Kombination mit einer 2-fachen Entwicklung. Koffein und andere Alkaloide wurden vollständig entfernt. LCMS/MS Kalibriergeraden von matrix-matched und Lösemittelstandards und sehr gute mittlere Wiederfindungen, bestimmt gegen Lösemittelstandards, belegten erneut die Leistungsfähigkeit der HTpSPE. Das neue Acetonitril-HTpSPE Verfahren ergab hinsichtlich aller Validierungsparameter zuverlässigere und exaktere Resultate als die QuEChERS-dSPE Methode.
In den letzten Jahren ist die allumfassende Analytik mittels Screening-Methoden in der Rückstandsanalytik von Pestiziden immer wichtiger geworden. Vor diesem Hintergrund wurde ein Screening-Konzept für HTpSPE-Extrakte entwickelt, das ein hochauflösendes MS nutzt, um die gereinigten Extrakte auf Pestizidrückstände zu untersuchen, ohne dass eine chromatographische Trennung notwendig ist. Diese Kopplung lieferte eine völlig neue Strategie für ein schnelles Screening, die direkte Mikroliter-Fließinjektionsanalyse mittels Flugzeitmassenspektrometrie (µL-FIATOFMS). Der neue HTpSPEµL-FIATOFMS Ansatz ermöglichte die simultane Erfassung aller Pestizide in einem einzigen Massenspektrum innerhalb von wenigen Minuten. Dank der HTpSPE waren die erhaltenen Spektren nahezu frei von Matrixverbindungen. Unter Verwendung der Intensitäten der Massensignale lieferte das HTpSPEµL-FIATOFMS Verfahren sehr gute mittlere Wiederfindungen gegen Lösemittelstandards für alle Matrices und Pestizide die Gegenstand dieser Studie waren. Im Vergleich zur dSPE zeigte die HTpSPE deutlich bessere Ergebnisse für alle getesteten Parameter. Mithilfe einer eigenen, neu entwickelten Datenbank-basierten Suchstrategie wurden alle dotierten Pestizide erkannt und korrekt identifiziert, während nahezu keine falsch-positiven Treffer auftraten. Zusätzlich wurde eine non-target Suchoption erfolgreich etabliert. Sie lieferte eine Liste an exakten Massen aller Probenbestandteile, die in den injizierten Extrakten vorhanden, aber nicht Bestandteil der Datenbank sind. Das neue HTpSPEµL-FIATOFMS Screening wurde schließlich erfolgreich auf diverse reale Proben angewandt. Die identifizierten Pestizide entsprachen denjenigen, die zuvor in einem Handelslabor mittels LCMS/MS Messungen von QuEChERS-dSPE Extrakten detektiert wurden.

Nowadays, the usage and partly abuse of veterinary antibiotics resulted in a very pressing need to control residues in foods of animal origin. Particularly, the increasingly demanding MRL issues and the huge number of samples to be monitored raised great challenges in this field. Microbial growth inhibition assays are traditionally employed for screening purposes, while sophisticated HPLC-MS methods are alternatively used or only used for confirmation purposes. To substitute the time consuming growth inhibition assays, HPTLC as a platform hyphenated to multi detection modes was employed in this study for the development of a high throughput, sensitive and cost-efficient screening-oriented methodology for antibiotics residues.
The first step was focused on tetracyclines and fluoroquinolones, which are the most problematic antibiotics in the European Union and account for the most of the used veterinary antibiotics. To prevent strong tailing effects, the separation was optimized on normal-phase silica gel plates modified with ethylenediamine tetraacetic acid (EDTA). Besides, selective and sensitive fluorescence densitometry was optimized to achieve best signal/noise ratios. Under these conditions, limits of detection (LODs) and quantitation (LOQs) were in the range 1225 and 4595 µg/kg, respectively. Recoveries from milk samples, spiked at 50, 100 and 150 µg/kg and extracted by a modified QuEChERS procedure, ranged from 76 to 105%. To circumvent the ion suppressions due to EDTA, HPTLC-mass spectrometry (HPTLC-MS) was optimized, allowing the selective confirmation of positive findings, also offering high sensitivity of 25 µg/kg, and meeting Commission Regulation (EU) No. 37/2010.
In the second step, sulfonamides were targeted, which are the secondly most administered veterinary antibiotics in the European Union. Separation of twelve most important sulfonamides was achieved on HPTLC silica gel plates, followed by fluram derivatization and sensitive and selective quantitation by fluorescent densitometry. LODs and LOQs were determined to 1540 and 3570 µg/kg, respectively. Samples of bovine milk, porcine liver and kidney were extracted according to the QuEChERS strategy. Additionally, a confirmative detection by HPTLC-MS was optimized, offering straightforward identification of target zones. The method was validated to meet the enforced Commission Regulation (EU) No. 37/2010.
Finally, a more universal screening method based on HPTLC-bioautography was developed for most of the first-line veterinary antibiotics. A comprehensive HPTLC plate test revealed that the bio-compatibility of different plate layer materials to the applied bioluminescent bacteria (A. fischeri DSM No. 7151) was surprisingly different. It was then discovered that both bright bioluminescent background and significant inhibition zones of antibiotics can only be achieved on HPTLC amino F254S plates.
In this case, HPTLC was not used for the chromatographic separation of individual antibiotics extracted with acetonitrile, but in terms of planar solid phase extraction to separate bioactive matrix compounds and to focus the analytes within two distinct target zones of different polarity. Together with HPTLC-MS for identification and confirmation purposes, the developed procedure enabled the rapid, sensitive and efficient multi-class screening of antibiotic residues (16 species of 5 groups, except sulfonamides and penicillins, which only affect Gram positive bacteria). The multi-sample plate images provided the results within a few hours. Thanks to the high sensitivity and the great matrix tolerance, the established method was successfully applied to bovine milk and porcine kidney samples, each spiked at the EU MLRs.
Der enorme Einsatz und partielle Missbrauch von Antibiotika in der Tiermedizin führte zu dringend notwendigen Maßnahmen, um Rückstände in tierischen Lebensmitteln zu kontrollieren. Insbesondere die steigend anspruchsvollen Rückstandshöchstgehalte sowie die geforderten zunehmenden Probenzahlen stellen eine große Herausforderung dar. Mikrobielle Hemmhof-Assays werden traditionell zum Proben-Screening eingesetzt, während anspruchsvolle HPLC-MS Methoden häufig alternativ oder nur zur Absicherung positiver Befunde verwendet werden. Um die zeitintensiven Hemmhof-Assays zu ersetzen, wurde in dieser Arbeit die HPTLC gekoppelt mit Multi-Detektionsmethoden eingesetzt, um eine schnelle und kostengünstige Screening-orientierte Methodik für Antibiotika-Rückstände zu entwickeln.
Der erste Teil der Arbeit konzentrierte sich auf zwei Gruppen von "schwierigen" Antibiotika, Tetracycline und Fluorchinolone, die zu den häufigst eingesetzten veterinärmedizinischen Antibiotika gehören. Unter Vermeidung von Tailing-Effekten wurde die HPTLC-Trennung auf Normalphasen-Kieselgelplatten, modifiziert mit Ethylendiamintetraessigsäure (EDTA), optimiert. Außerdem wurde eine selektive und empfindliche Fluoreszenz-Densitometrie genutzt, um beste Signal/Rausch-Verhältnisse zu erreichen. Nachweis- und Bestimmungsgrenzen lagen im Bereich von 12-25 und 45-95 µg/kg. Wiederfindungen aus Milchproben, dotiert auf 50, 100 und 150 µg/kg und extrahiert mit einer modifizierten QuEChERS-Methode, ergaben sich zu 76-105%. Zur Absicherung positiver Befunde wurde die Massenspektrometrie (HPTLC-MS) bezüglich Ionensuppressionen durch EDTA dahingehend optimiert, dass eine empfindliche Detektion von 25 µg/kg möglich war und die Vorgaben der Verordnung (EU) Nr. 37/2010 erfüllt wurden.
Im zweiten Schritt galt das Interesse den Sulfonamiden, den zweithäufigst eingesetzten veterinärmedizinischen Antibiotika. Die Trennung erfolgte auf HPTLC-Kieselgelschichten und nachfolgender Fluram-Derivatisierung. Dies erlaubte eine selektive und sehr sensitive Quantifizierung der zwölf bedeutendsten Sulfonamide durch Fluoreszenz-Densitometrie. Nachweis- und Bestimmungsgrenzen ergaben sich zu 15-40 und 35-70 µg/kg. Die Extraktion von Milch-, Leber- und Nierenproben erfolgte mit Acetonitril (analog "QuEChERS"). Zur Bestätigung positiver Proben wurde erneut die HPTLC-MS Kopplung zur einfachen Identifizierung der Zielzonen optimiert. Die Methode wurde hinsichtlich der Vorgaben der Verordnung (EU) Nr. 37/2010 für Milch sowie Schweine-Leber und -Nieren validiert.
Schließlich wurde eine universelle Screening-Methode mittels HPTLC-Bioautographie für die meisten der First-Line Tier-Antibiotika entwickelt. Ein umfangreicher HPTLC-Plattentest zeigte, dass die Bio-Kompatibilität der verschiedenen Schichtmaterialien mit den eingesetzten Leuchtbakterien (Aliivibrio fischeri DSM-Nr. 7151) überraschend unterschiedlich war. Nur auf HPTLC-Amino F254S Platten zeigten die Bakterien die optimale Biolumineszenz und damit signifikante Hemmzonen für Antibiotika.
Nach Extraktion mit Acetonitril wurde die HPTLC hier nicht zur Auftrennung der einzelnen Antibiotika optimiert, sondern im Sinne einer planaren Festphasenextraktion zur Abtrennung ebenfalls bioaktiver Matrixkomponenten und zur Fokussierung der Antibiotika in zwei unterschiedlich polare Zielzonen. Zusammen mit der HPTLC-MS Kopplung lieferte diese Methode ein schnelles, empfindliches und effizientes Multi-Class-Screening von Antibiotika-Rückständen (16 Wirkstoffe aus 5 Gruppen, außer Sulfonamide und Penicilline, welche nur Gram-positive Bakterien hemmen). Ein Plattenbild lieferte innerhalb weniger Stunden das Ergebnis für viele Proben. Dank hoher Empfindlichkeit und großer Matrixtoleranz wurde die Methode erfolgreich auf Milch- und Schweinenieren-Proben angewendet, dotiert auf die EU-Rückstandshöchstgehalte.

Although UV filters are important, widespread used cosmetic ingredients, their reaction potential towards skin proteins has hardly been studied so far. Therefore, the aim of the present thesis was to investigate the reactivity of widespread UV filter substances towards skin proteins using increasingly complex protein and skin model systems and different analytical techniques.
At first, the development of a rapid high-performance thin-layer chromatographic (HPTLC) screening method on an amino phase as protein model provided an easy and rapid way to estimate the reactivity of the common UV filters benzophenone-3 (BP-3), hydroxymethoxybenzoyl sulfonic acid (HMBS), butyl methoxydibenzoylmethane (BM-DBM), 3-benzylidene camphor (3 BC), 4 methylbenzylidene camphor (4 BMC), octocrylene (OCR), ethylhexyl methoxycinnamate (EHMC), ethylhexyl salicylate (EHS), diethylhexyl butamido triazone (DEBT), ethylhexyl triazone (EHT), and octyldimethyl p-aminobenzoic acid (OD-PABA) towards amino groups under thermal and irradiation conditions. A direct comparison of the results of the screening with (photo) patch test data of the dermatological practice showed that especially those UV filters, which are known to be common triggers for (photo) allergic reactions, showed the highest tendency to bind to the amino phase. This indicates that the screening may be well suited to identify possible skin sensitizers as part of a multistage testing strategy.
The observation that the reactivity of the different UV filters was influenced by both heat and UV irradiation was verified during the subsequent studies with butylamine and ethanolamine. The UV filters showed individual, time- and temperature-dependent reactivities towards the amines. Benzophenone imines, enamines, and amides were identified as typical reaction products by means of electrospray ionization mass spectrometry (ESI-MS), Fourier transform infrared (FTIR), and nuclear magnetic resonance (NMR) spectroscopy. BP-3, HMBS, the dibenzoylmethanes, OCR, and EHS showed by far the highest reactivity what was in good correlation with the previous screening, indicating a different contact-allergic potential of the UV filters. In contrast, the esters EHMC and EHT showed a significantly lower reactivity, and for the UV filters 3 BC, 4-MBC, and OD-PABA no conversion was observable at all.
The formation of the reaction products had partly big influence on the respective UV filter spectra. In the case of BP-3, HMBS, and EHS, the conversions led to a strong bathochromic shift and hence to approved UVA protection. In contrary, in the case of DBM and BM-DBM and especially OCR, a breakage of the original molecule structures was observed, resulting in a significant decrease of the respective absorption strength.
The same reaction tendencies could also be observed, when using Boc-protected lysine, the tetrapeptide Boc-Gly-Phe-Gly-Lys-OH (Boc-GFGK), and bovine serum albumin (BSA) as increasingly complex protein or skin models. OCR and BM-DBM confirmed to be most reactive towards the lysine side chains of the mentioned model systems, followed by DBM > BP 3 > EHS > EHMC > EHT in decreasing order.
To determine the covalent binding of the UV filters to the protein BSA, beside the extraction of the unbound UV filters, the increase of the molar mass of the formed BSA-adducts was additionally exemplarily determined for EHMC and DBM by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS). Both methods gave comparable results. Binding to BSA did not affect the UV absorption properties of BM-DBM, EHMC, and EHT, but led to a bathochromic shift in the cases of BP-3 and EHS. For OCR, a strong hypsochromic shift and a nearly complete loss of UVA+B protection was observable.
To better reflect the usual application conditions, a thin gelatine layer was chosen as further skin model. The UV filter amounts applied were adapted according to the existing ISO norm for the determination of the SPF. Afterwards, UV irradiation was performed. The binding amounts were determined both by extraction of the unbound UV filters and by isotope-ratio mass spectrometry (IRMS), where two synthetized, stable-isotope labelled UV filter analogues (EHC-d5 and DBM-d5) were used. In contrast to the esters EHMC and EHT, which showed comparatively small binding amounts, for the UV filters OCR, BP-3, EHS, and BM-DBM significant reaction tendencies towards gelatin were observed.
Finally, various commercial sunscreens and personal care products with UV protection were applied on either prepared porcine skin or glass plates, followed by UV irradiation. Significant differences were observed for the amounts of UV filters extracted from glass and skin. The lower recoveries in the case of the skin indicated an additional reaction of the UV filters towards the skin samples. BP-3 showed the highest discrepancy between the recoveries from glass and skin after irradiation, followed by EHS > BM DBM > OCR > EHMC > EHT in decreasing order.
The present dissertation showed that cosmetic UV filters were able to react with amino structures of different proteins under thermal and irradiation conditions. As the formation of protein adducts is seen as key event in the development of (photo) allergic reactions, the results indicate a specific skin sensitization potential of the UV filters. This is confirmed by the experience of dermatological practice. Since such reactions have partly strong influence on the respective UV filter spectra, the existing in vitro methods using PMMA or quartz glass as substrates have to be questioned, since those methods cannot capture such skin-typical reactions.
Obwohl UV-Filter vielfach verwendete kosmetische Inhaltsstoffe sind, wurde ihr Reaktionsvermögen gegenüber Hautproteinen bisher kaum untersucht. Das Ziel der vorliegenden Arbeit war es, die Reaktivität weit verbreiteter UV-Filters gegenüber Hautproteinen zu untersuchen. Hierfür wurden unterschiedliche, komplexer werdende Protein- und Hautmodelle sowie unterschiedliche analytische Methoden herangezogen.
Im ersten Schritt wurde eine schnelle hochleistungsdünnschichtchromatographische (HPTLC) Screening-Methode auf Basis einer Aminophase als Proteinmodell entwickelt. Durch die Bestimmung des Bindungsvermögens der UV-Filtersubstanzen Benzophenon-3 (BP-3), 2-Hydroxy-4-methoxybenzophenon-5-sulfonsäure (HMBS), Octocrilen (OCR), Butylmethoxydibenzoylmethan (BM-DBM), Dibenzoylmethan (DBM), Ethylhexylsalicylsäure (EHS), 3-Benzylidencampher (3-BC), 4-Methylbenzylidencampher (4-MBC), Ethylhexylmethoxyzimtsäure (EHMC), Diethylhexylbutamidotriazon (DEBT), Ethylhexyltriazon (EHT) und Octyldimethyl-p-aminobenzoesäure (OD PABA) an die Aminoschicht konnte die generelle Reaktivität der UV-Filter gegenüber Proteinstrukturen unter dem Einfluss von Wärme und UV-Bestrahlung bestimmt werden.
Ein direkter Vergleich der Ergebnisse des Screenings mit (Photo)patchtest-Daten aus der dermatologischen Praxis zeigte, dass insbesondere die UV-Filter, die als typische Auslöser (photo)allergischer Reaktionen gelten, auch auf der Aminophase die größte Bindungstendenz zeigten. Dies legt die Vermutung nahe, dass das entwickelte Screening als Teil einer Testbatterie dazu beitragen kann, mögliche sensibilisierende Stoffe zu identifizieren.
Die Reaktivität der untersuchten UV-Filter gegenüber Aminstrukturen wurde anschließend durch in vitro Untersuchungen mit Butylamin und Ethanolamin überprüft. Die UV-Filter zeigten eine zeit- und temperaturabhängige Reaktivität gegenüber den beiden Aminen. Mittels Elektrospray-Ionisations-Massenspektrometrie (ESI-MS), Infrarotspektroskopie (IR) und Kernspinresonanzspektroskopie (NMR) konnten Benzophenonimine, Enamine und Amide als typische Reaktionsprodukte identifiziert werden. BP-3, HMBS, DBM, BM DBM, OCR und EHS zeigten bei weitem die größte Reaktivität. EHMC und EHT hingegen zeigten eine deutlich geringere Reaktivität und für 3-BC, 4-MBC und OD PABA konnte keinerlei Umsatz detektiert werden.
Die Bildung der unterschiedlichen Reaktionsprodukte führte teils zu starken Veränderungen im Absorptionsspektrum der UV-Filter. Für BP-3, HMBS und EHS wurde eine bathochrome Verschiebung der Spektren und damit eine erhöhte UVA-Schutzwirkung festgestellt. Im Gegensatz dazu war im Falle von DBM, BM-DBM und OCR ein Bruch der ursprünglichen Struktur feststellbar, was zu einem deutlichen Verlust der Absorptionsstärke führte.
Ähnliche Reaktionstendenzen der einzelnen UV-Filter waren auch für die komplexer werdenden Reaktionspartner Boc-Lysin, das Tetrapeptid Boc-Gly-Phe-Gly-Lys-OH (Boc-GFGK) und Rinderserumalbumin (BSA) feststellbar. OCR und BM-DBM zeigten die größte Reaktivität, gefolgt von den UV-Filtern DBM > BP 3 > EHS > EHMC > EHT in abnehmender Reihenfolge.
Um eine Bindung der UV-Filter an BSA festzustellen, wurde zusätzlich zur Extraktion der ungebundenen UV-Filter die Veränderung der molaren Masse des Proteins beispielhaft für EHMC und DBM mittels Matrix-unterstützter Laser-Desorption/Ionisation (MALDI-TOFMS) bestimmt. Beide Methoden zeigten vergleichbare Ergebnisse. Im Falle von BM DBM, EHMC und EHT zeigten sich durch die Bindung an BSA keine deutlichen Unterschiede in den UV-Spektren, bei EHS und BP-3 war hingegen eine bathochrome Verschiebung zu beobachten. Für OCR zeigte sich eine hypsochromen Verschiebung und ein fast vollständiger Verlust der UVB-Schutzwirkung.
Um die realen Anwendungsbedingungen noch besser abzubilden, wurde eine dünne Gelatineschicht als Hautmodell verwendet und nach Auftragung der UV-Filter bestrahlt. Die Bindungsmenge der UV Filter wurde mittels Extraktion der ungebundenen Substanzen sowie Isotopenverhältnis-Massenspektrometrie (IRMS) unter Verwendung der synthetisierten, Deuterium-markierten UV Filteranaloga EHC-d5 und DBM-d5 bestimmt. Im Gegensatz zu den Estern EHMC und EHT, für die eher geringe Bindungsmengen festgestellt wurden, zeigten OCR, BP-3, EHS und BM-DBM eine deutliche Bindungstendenz gegenüber der Gelatineschicht.
Abschließend wurden unterschiedliche handelsübliche Sonnenschutzprodukte auf präparierte Schweinehaut sowie Glasplättchen aufgetragen und bestrahlt. Bei der sich anschließenden Extraktion der UV-Filter wurden signifikante Unterschiede zwischen den jeweils ermittelten UV-Filtergehalten festgestellt. Die geringeren Wiederfindungen im Falle der Schweinehaut ließen auf eine zusätzliche Reaktion der UV-Filter auf der Haut schließen. Für BP-3 war der deutlichste Unterschied zwischen Glas und Haut feststellbar, gefolgt von EHS > BM-DBM > OCR > EHMC > EHT in absteigender Reihenfolge.
Mit der vorliegenden Dissertation konnte gezeigt werden, dass kosmetische UV-Filter unter dem Einfluss von Wärme und UV-Licht in der Lage sind, mit Aminostrukturen unterschiedlicher Proteine zu reagieren. Da die Bildung von Proteinaddukten als entscheidender Schritt bei der Ausbildung von (Photo)allergien gilt, lassen die Ergebnisse auf ein Sensibilisierungspotential der Inhaltsstoffe schließen. Dies wird durch die Erfahrungen aus der dermatologischen Praxis bestätigt. Da sich die Reaktionen teils stark auf die UV-Spektren der jeweiligen UV-Filter und deren Schutzleistung auswirken, sollten die existierenden in vitro Methoden zur Bestimmung der Wirksamkeit von Sonnenschutzprodukten kritisch hinterfragt werden, da beim Einsatz inerter Substratmaterialien wie PMMA oder Quarzglas der Einfluss hautspezifischer Reaktionen nicht berücksichtigt wird.

Substances that disrupt or impair the hormone system (endocrine system) or that show an irreversible influence on it are referred to as endocrine disruptors or xenohormones. Concerning this, also estrogen active compounds (EAC) are endocrine disruptors, that are under suspicion of being involved in the formation of tumors or to induce disruption during development and reproduction, and are, for example, blamed for being responsible for the feminization of fish. At this, EAC can be natural (human, phytoestrogens) but also synthetic substances, which are discharged to the environment by humans (e.g. pharmaceuticals, pesticides, additives). Regarding the ubiquitous presence of EAC, suitable methods for the analysis of EAC are required. An in vitro method for the determination of EAC is the YES assay (yeast estrogen screen) that is executed in liquid solutions in microtiter plates and that works with genetically modified yeasts, which contain the human estrogen receptor (hER) and a reporter gene encoding for the enzyme beta-galactosidase. In presence of EAC, the enzyme is produced and subsequently cleaves a substrate that is used to measure the receptor activity and thus the estrogenic activity.
The transfer of the YES assay to high-performance thin-layer chromatography (HPTLC) was successfully demonstrated and advanced, thus resulting in the combination of a chromatographic separation of analytes and the detection of EAC using genetically modified yeast cells directly on the HPTLC plate (HPTLC planar yeast estrogen screen, HPTLC-pYES). Usually, the substrate 4-methylumbelliferyl-beta-D-galactopyranoside is used for pYES, releasing blue fluorescing 4-methylumbelliferone (MU) after enzymatic cleavage. Various matrices, however, often contain a plenty of different components, partly showing native fluorescences (blue, red), why the detection of the blue fluorescing MU can be interfered.
By applying the substrate resorufin-beta-D-galactopyranoside (RGP) and by using automated devices, the RGP-pYES as fast screening tool for EAC was developed and successfully applied to waste water samples and extracts of hops pellet samples. A screening method using HPTLC simultaneously represents a planar clean-up, why samples do not have to undergo complex steps of sample preparation or purification. The chromatographic separation in combination with the detection of estrogenic activity using genetically modified yeasts directly on the plate allowed the detection, the determination and the identification of single EAC. Using RGP, which releases orange fluorescing resorufin after enzymatic cleavage as positive signal of estrogenic activity, enabled a clear differentiation between fluorescences due to estrogenicity and the native fluorescence of sample components. Application of the RGP-pYES to spiked water samples and sewage samples showed high recovery rates and a good precision, and thus the applicability of the method as screening tool for environmental samples.
By means of suitable evaluation methods, additionally the generation of dose-response curves of known and unknown EAC and thus the generation of so-called logit-log plots was possible. This enabled the determination of estradiol equivalent factors of known EAC as well as the determination of estradiol equivalent concentrations and amounts, respectively, of known and unknown EAC in liquid and solid samples. Thus, the possibility to estimate the estrogenic potential of a sample or single sample components was given. The coupling of pYES to mass spectrometry additionally allowed the identification of unknown EAC, demonstrated exemplarily by investigation of extracts of hops pellet samples, in which the only detected EAC in the hops extracts was identified as prenylnaringenin.
Since the method uses a planar system, the pYES advantageously reveals all chromatographically separated sample components at one look and, as bioassay, additionally detects a possible estrogenic activity (activity at the hER) of single substances, while a differentiation between native occurring fluorescences of sample contaminants and the fluorescence as positive signal for estrogenicity of a substance is granted.
Stoffe, welche das Hormonsystem (endokrine System) irreversibel beeinflussen, stören oder beeinträchtigen, werden als endokrine Disruptoren oder auch als Xenohormone bezeichnet. Hierzu zählen ebenso estrogen wirksame Substanzen (estrogen active compounds, EAC), welche unter anderem im Verdacht stehen bei der Tumorbildung beteiligt zu sein oder Störungen in der Entwicklung und Reproduktion hervorzurufen, und beispielsweise für die Verweiblichung von Fischen verantwortlich zu sein. EAC können hierbei natürlichen Ursprungs (menschlich, pflanzlich) sein oder synthetisch hergestellte Stoffe, welche durch den Menschen in die Umwelt eingetragen werden (z.B. Arzneimittel, Pestizide, Additive). Aufgrund ihres ubiquitären Vorkommens sind geeignete Methoden zur Untersuchung auf EAC erforderlich. Eine in vitro Methode zur Bestimmung von EAC ist der YES-Assay (yeast estrogen screen), welcher in flüssigen Medien in Mikrotiterplatten durchgeführt wird und mit genetisch modifizierten Hefen arbeitet, die den humanen Estrogenrezeptor (human estrogen receptor, hER) und ein Reportergen enthalten, welches für das Enzym beta-Galactosidase kodiert. In Anwesenheit von EAC wird das Enzym produziert und setzt anschließend ein Substrat um, welches zur Messung der Rezeptor- und damit der Estrogenaktivität dient.
Die Übertragung des YES-Assay auf die Hochleistungs-Dünnschichtchromatographie (high-performance thin-layer chromatography, HPTLC) wurde erfolgreich gezeigt und weiterentwickelt, so dass eine chromatographische Trennung der Analyten mit dem Nachweis einer möglichen estrogenen Aktivität unter Verwendung genmodifizierter Hefezellen direkt auf der HPTLC-Platte kombiniert werden konnte (HPTLC planar yeast estrogen screen, HPTLC-pYES). Für gewöhnlich wird das Substrat 4-Methylumbelliferyl-beta-D-Galactopyranosid für den pYES eingesetzt, welches nach enzymatischer Spaltung blau fluoreszierendes 4-Methylumbelliferon (MU) freisetzt. Häufig beinhalten unterschiedliche Matrices jedoch viele verschiedene Begleitkomponenten, welche teilweise native Fluoreszenzen aufzeigen (blau, rot), wodurch der Nachweis des blau fluoreszierenden MU gestört werden kann.
Durch die Wahl des Substrates Resorufin-beta-D-Galactopyranosid (RGP) und unter Verwendung automatisierter Geräte wurde der RGP-pYES als schnelles Screening auf EAC entwickelt und erfolgreich auf Abwasserproben und Extrakte von Hopfenpellets angewandt. Ein Screening unter Einsatz der HPTLC stellt gleichzeitig ein planares Clean-up dar, wodurch die Proben keinen aufwändigen Reinigungs- oder Aufarbeitungsschritten unterzogen werden müssen. Die chromatographische Trennung in Kombination mit dem Nachweis estrogener Aktivität mittels genmodifizierter Hefen direkt auf den Platten ermöglichte den Nachweis, die Bestimmung und die Identifizierung einzelner EAC. Die Verwendung des RGP, welches durch enzymatische Spaltung orange fluoreszierendes Resorufin als positiven Nachweis estrogener Aktivität freisetzt, erlaubte hierbei eine klare Differenzierung von Fluoreszenzen aufgrund estrogener Wirksamkeit gegenüber der nativen Fluoreszenz von Probenkomponenten. Die Anwendung des RGP-pYES auf Extrakte gespikter Wasserproben und Abwasserproben zeigte hohe Wiederfindungsraten und eine gute Präzision und damit die Eignung der Methode als Screening-Tool für Umweltproben.
Mittels geeigneter Auswerteverfahren war es zudem möglich, Dosis-Wirkungs-Kurven bekannter und unbekannter EAC und daraus sogenannte logit-log Plots zu erstellen. Hiermit war die Bestimmung von Estradiol-Äquivalenz-Faktoren bekannter EAC sowie die Ermittlung von Estradiol-Äquivalenz-Konzentrationen bzw. -Gehalten bekannter und unbekannter EAC in flüssigen und festen Proben möglich. Damit war die Möglichkeit der Abschätzung des estrogenen Potentials einer Probe oder einzelner Probenkomponenten gegeben. Die Kopplung des pYES mit der Massenspektrometrie erlaubte zusätzlich die Identifizierung unbekannter EAC, wie am Beispiel der Untersuchung von Extrakten von Hopfenpellets gezeigt werden konnte, in welchen Prenylnaringenin als einzig nachweisbare EAC der Hopfenextrakte identifiziert wurde.
Als Methode im planaren System bietet der pYES den Vorteil, alle chromatographisch getrennten Probenbestandteile auf einen Blick zu sehen und zusätzlich als Bioassay eine mögliche estrogene Aktivität (Aktivität am hER) einzelner Substanzen nachzuweisen, während zwischen nativ auftretenden Fluoreszenzen von Probenkomponenten und der Fluoreszenz als Positivsignal der Estrogenaktivität einer Substanz differenziert werden kann.

Die Strahlentherapie stellt eine zentrale Säule bei der Behandlung von Krebs dar. Die maximale Dosis, mit der ein Tumor bestrahlt werden kann, wird jedoch durch Nebenwirkungen des mitbestrahlten Normalgewebes begrenzt. Um den Behandlungserfolg zu verbessern, ist es somit von wesentlichem Interesse, neue Therapiestrategien zu entwickeln, die gezielt die zellschädigende Wirkung der Strahlung auf den Tumor verstärken, während das gesunde Gewebe geschont wird. Dabei gilt es, Unterschiede zwischen Tumorzellen und normalen Zellen zu nutzen. So ist es ein charakteristisches Merkmal von Tumorzellen, Glukose unabhängig von der vorherrschenden Sauerstoffkonzentration überwiegend zu Laktat zu verstoffwechseln (Warburg-Effekt, aerobe Glykolyse), während normale Zellen bei Anwesenheit von Sauerstoff die meiste Glukose in den Mitochondrien oxidieren. Da der Warburg-Effekt im Vergleich zur mitochondrialen Glukoseoxidation jedoch mit einer geringen ATP-Ausbeute pro Molekül Glukose einhergeht, sind Tumorzellen auf eine hohe Glukosezufuhr angewiesen. Daher war es Ziel dieser Arbeit zu untersuchen, ob ein Glukoseentzug im Rahmen der Strahlentherapie, die eine energieaufwendige Reparatur von DNA-Schäden erfordert, eine geeignete Strategie darstellt, um selektiv die Strahlenempfindlichkeit von Tumorzellen, nicht jedoch von normalen Zellen, zu erhöhen. Es zeigte sich, dass ein Glukoseentzug die Proliferation der Tumorzelllinien A549 und FaDu, nicht jedoch der normalen Fibroblasten HSF7, hemmte. Zudem führte der Entzug von Glukose selektiv bei den Tumorzellen zum Zelltod, wobei dieser hauptsächlich durch Nekrose erfolgte. Wurde der Glukoseentzug mit einer Strahlenbehandlung kombiniert, so konnte eine selektive Radiosensitivierung der beiden Tumorzelllinien erzielt werden, welche mit einer beeinträchtigten Reparatur strahleninduzierter DNA-Doppelstrangbrüche (DNA-DSBs) einherging. Dabei stellte sich heraus, dass die Glukose in Tumorzellen für die späte Phase der DNA-DSB-Reparatur, beginnend ab 13 h nach der Bestrahlung, essentiell ist. Des Weiteren konnte in den Tumorzellen eine Hemmung der strahleninduzierten Histonacetylierung sowie KAP1-Phosphorylierung infolge des Glukoseentzugs beobachtet werden, welche auf eine Beeinträchtigung der strahleninduzierten Chromatinrelaxation schließen ließ. Da die Öffnung der Chromatinstruktur besonders für die Reparatur von DNA-DSBs innerhalb des Heterochromatins von Bedeutung ist und gerade diese DSBs zu späten Zeitpunkten nach der Bestrahlung repariert werden, ist zu vermuten, dass in Tumorzellen durch den Glukoseentzug vor allem die Reparatur heterochromatischer DNA-DSBs beeinträchtigt wird. Weitere Untersuchungen ergaben, dass ein Glukoseentzug in Tumorzellen keinen Mangel an nukleärem Acetyl-CoA, dem Substrat für die Acetylierung von Histonen, verursacht, weshalb dies als Grund für die beobachtete Hemmung der Histonacetylierung ausgeschlossen werden konnte. Es ist jedoch bekannt, dass die Histondeacetylase Sirt1 infolge eines Glukoseentzugs aktiviert wird. Die Sirt1-vermittelte Histondeacetylierung könnte der strahleninduzierten Histonacetylierung entgegenwirken und somit die Chromatinrelaxation und damit auch die Reparatur von DNA-DSBs nach der Bestrahlung beeinträchtigen. Tatsächlich konnte gezeigt werden, dass eine Hemmung von Sirt1 mittels Sirtinol die unter glukosefreien Bedingungen in Tumorzellen auftretende Beeinträchtigung der Reparatur strahleninduzierter DNA-DSBs teilweise aufheben kann. Der hemmende Effekt eines Glukoseentzugs auf die DNA-DSB-Reparatur konnte in Tumorzellen jedoch nicht nur unter glukosefreien Bedingungen beobachtet werden. So genügte eine Reduktion der Glukosekonzentration auf 0,5 g/l, um die DSB-Reparatur nach der Bestrahlung in gleichem Maße zu beeinträchtigen wie durch einen kompletten Entzug der Glukose. Des Weiteren stellte sich heraus, dass die Reparatur von DNA-DSBs in Tumorzellen unter glukosefreien Bedingungen bereits nach einer Bestrahlung mit 2 Gy durch die Autophagie begünstigt wird. Schließlich wurde gezeigt, dass neben der DNA-DSB-Reparatur auch der Stoffwechsel der Tumorzellen durch den Glukoseentzug beeinflusst wird. So beeinträchtigte der Entzug von Glukose die strahleninduzierte Umstellung des Glukosestoffwechsels, die mit einer Verstärkung der aeroben Glykolyse und einer Abnahme der mitochondrialen Glukoseoxidation einherging, was ebenfalls zur Radiosensitivierung der Zellen beitragen kann. Im Gegensatz zu den Tumorzellen führte der Glukoseentzug bei den normalen Fibroblasten weder zu einer Radiosensitivierung noch zu einer Beeinträchtigung der Reparatur strahleninduzierter DNA-DSBs. Zudem hatte der Glukoseentzug hier keinen Einfluss auf die Histonacetylierung sowie KAP1-Phosphorylierung nach der Bestrahlung. Diese Ergebnisse zeigen, dass ein Glukoseentzug in vitro eine geeignete Strategie darstellt, um selektiv Tumorzellen gegenüber einer Strahlenbehandlung zu sensitivieren, ohne die Strahlenempfindlichkeit normaler Zellen zu beeinflussen.
Radiotherapy is a major pillar of cancer treatment. However, the maximal dose that can be applied to a tumor is limited by side-effects of the irradiated normal tissue. Therefore, to improve treatment success, it is of significant interest to develop new treatment strategies that selectively enhance the cytotoxic effect of radiation in tumor cells while sparing healthy tissue. For this purpose, it is necessary to exploit differences between tumor cells and normal cells. Thus, tumor cells are characterized by metabolizing glucose preferentially to lactate regardless of the availability of oxygen (Warburg effect, aerobic glycolysis), while normal cells oxidize most of the glucose in the mitochondria if oxygen is present. Because the Warburg effect only produces low amounts of ATP per molecule of glucose when compared to mitochondrial glucose oxidation, tumor cells rely on high glucose supply. Hence, it was the aim of this study to investigate whether a glucose starvation during radiotherapy, which requires energy-dependent repair of DNA damage, is an appropriate strategy to selectively enhance radiosensitivity of tumor cells, but not of normal cells. It was shown that glucose starvation inhibited proliferation of the tumor cell lines A549 and FaDu, but not that of the normal fibroblasts HSF7. Moreover, deprivation of glucose induced cell death selectively in tumor cells, which occurred mainly via necrosis. Combining glucose starvation with radiotherapy led to selective radiosensitization of both tumor cell lines, which was accompanied by impaired repair of radiation-induced DNA double-strand breaks (DNA DSBs). In this context, it turned out that in tumor cells glucose is essential for the late stage of DNA DSB repair starting from 13 h after irradiation. Furthermore, an inhibition of radiation-induced histone acetylation as well as KAP1 phosphorylation could be observed in tumor cells following glucose starvation, indicating an impairment of radiation-induced chromatin relaxation. Because opening of the chromatin structure is particularly important for the repair of DNA DSBs within heterochromatin and because these DSBs are the ones that are repaired at late time points after irradiation, it can be assumed that in tumor cells glucose starvation mainly impairs the repair of heterochromatic DNA DSBs. Further investigations revealed that in tumor cells glucose starvation does not cause lack of nuclear acetyl-CoA, which is the substrate for the acetylation of histones, and therefore this could be excluded as cause of the observed inhibition of histone acetylation. However, it is known that the histone deacetylase Sirt1 is activated in response to glucose starvation. Histone deacetylation by Sirt1 could counteract radiation-induced histone acetylation, thus impairing chromatin relaxation as well as repair of DNA DSBs after irradiation. In fact, it was shown that inhibition of Sirt1 by sirtinol can partly abrogate the impaired repair of radiation-induced DNA DSBs that was observed in tumor cells under glucose-free conditions. However, the inhibitory effect of glucose starvation on DNA DSB repair in tumor cells could not only be observed under glucose-free conditions. Thus, reducing the glucose concentration to 0.5 g/l was enough to impair DSB repair following irradiation to the same degree as after total deprivation of glucose. Furthermore, it turned out that under glucose-free conditions DNA DSB repair in tumor cells was promoted by autophagy already after irradiation with 2 Gy. Finally, it was shown that, in addition to DNA DSB repair, also tumor metabolism is influenced by glucose starvation. Thus, deprivation of glucose impaired the radiation-induced switch of glucose metabolism that was characterized by increased aerobic glycolysis and decreased mitochondrial glucose oxidation, and this can also contribute to radiosensitization of the cells. In contrast to tumor cells, glucose starvation neither caused radiosensitization nor impaired the repair of radiation-induced DNA DSBs in normal fibroblasts. Moreover, in these cells, glucose starvation had no influence on histone acetylation and KAP1 phosphorylation after irradiation. These results demonstrate that glucose starvation is an appropriate in vitro strategy to selectively sensitize tumor cells to radiotherapy without influencing the radiosensitivity of normal cells.

Vitamin E is a generic term for a group of micronutrients exhibiting the biological activity of alpha-tocopherol. Initially, four tocopherols (T) and four tocotrienols (T3) were recognized as the naturally occurring vitamin E compounds. The main difference among T and T3 is the 3-fold unsaturated 16-carbon side chain of the T3 compared to the saturated 16-carbon side chain of the T. Recently, a group of four vitamin E compounds with a single double bond at carbon 11 were discovered, namely tocomonoenols (T1). Edible oils are the major source of T, T3, and of T1.
As a fat-soluble vitamin, the vitamin E is absorbed after oral intake and transported in the circulation to the liver, where vitamin E undergoes sorting by the action of the alpha-hepatic-tocopherol transfer protein (TTP) and the cytochrome P450 (CYP) enzymes. alpha-T is preferentially secreted into the bloodstream, while the non-alpha-T congeners are metabolized by CYP to the carboxyethylhydroxychromanols (CEHC), which are excreted via urine and feces. The TTP has been recognized as necessary for the maintenance of normal alpha-T concentrations in plasma and extrahepatic tissues. Interestingly, TTP might also protect the non-alpha-T congeners from side-chain degradation, and therefore prevent their metabolic degradation.
The present thesis aimed at increasing our knowledge of the non-αT congeners of vitamin E with respect to their occurrence in food, their intracellular localization upon uptake into liver cells, and their tissue distribution in mammals. A potential role of the TTP in the intracellular and intra-organismic trafficking of the non-alpha-T congeners was a second focus of the current investigations.
To this purpose, the vitamin E profiles and contents in oils of three Elaeis Guineensis, two Elaeis Oleifera, and one hybrid OxG palm fruit genotypes from Costa Rica were determined after mechanical extraction with a screw press and chemical extraction with hexane. Vitamin E profiles in the palm oils were similar, irrespective of the genotype and extraction procedure, and alpha- and gamma-T3 were the most abundant congeners. alpha-T1 was found in oils from five of the six varieties. Hexane extraction yielded up to 2.5-fold higher total vitamin E compared to screw press extraction.
The two most abundant tocotrienols in the oils were selected for further studies with respect to their cellular uptake and intracellular distribution in cultured liver cells with and without stable expression of TTP and compared to their respective tocopherol counterparts. After uptake, all four congeners were primarily associated with the lysosomes, endoplasmic reticulum and plasma membrane. Overall, the results conclude that neither the structural differences between the four congeners, nor the TTP-expression are important factors behind the intracellular trafficking (uptake and distribution) of the congeners in cultured liver cells.
Finally, an animal study was performed to examine the tissue distribution of alpha-T1 in mice in comparison to alpha-T. Besides was investigated the influence of TTP. Wild-type and TTP knockout mice were fed a standard diet with either alpha-T or alpha-T1 for 2 weeks. Concentrations were measured in blood and several tissues. alpha-T1 was only found in blood, not in tissues. Loss of TTP function in knockout mice resulted in almost complete depletion of alpha-T in all tissues. Interestingly, alpha-T1 was still present in blood. In conclusion, alpha-T1 reached the blood in mice with and without TTP function, suggesting that TTP may not, or only to a limited extent, be required for the secretion of alpha-T1 into the systemic circulation.
Since more is known about alpha-T than the non-alpha-T congeners, new opportunities for further research on the biological activities and consequent health benefits of the non-alpha-T congeners have arisen based on the contributions of the present thesis.
Vitamin E ist der Oberbegriff für eine Gruppe von Mikronährstoffen, welche die biologische Aktivität von alpha-Tocopherol aufweisen. Bisher wurden acht Kongenere, vier Tocopherole (T) und vier Tocotrienole (T3) als die natürlich vorkommenden Vitamin-E-Verbindungen klassifiziert. Die Unterscheidung zwischen T und T3 wird durch die dreifach ungesättigte 16-Kohlenstoff lange Seitenkette der T3´s im Vergleich zur gesättigten 16- Kohlenstoff langen Seitenkette der T´s definiert. Vor kurzem wurde eine Gruppe von vier Vitamin-E-Verbindungen mit einer Einfach-Doppelbindung am 11ten Kohlenstoffatom entdeckt und als Tocomonoenole (T1) bezeichnet. Speiseöle sind die Hauptquelle für T, T3 und T1.
Das fettlösliches Vitamin E wird oral aufgenommen und zur Leber transportiert. In der Leber durchlaufen die Vitamin E Kongenere eine Sortierung durch das alpha-hepatischen-Tocopherol-Transferprotein (TTP) und der Cytochrom P450 (CYP)-Enzyme. Das alphaT wird bevorzugt in den Blutkreislauf abgegeben, während die nicht-alphaT-Kongenere von den CYP-Enzymen zu den Carboxyethylhydroxychromanolen (CEHC) metabolisiert und über Urin und Stuhl ausgeschieden werden. Für die Aufrechterhaltung der normalen alphaT-Konzentration in Plasma und extrahepatischem Gewebe ist das TTP daher notwendig. Interessanterweise ist es möglich, das TTP auch die nicht-alphaT-Kongenere vor dem Abbau der Seitenkette schützen und so deren metabolischen Abbau verhindern kann.
Ziel dieser Arbeit war unser Wissen über die nicht-alphaT-Kongenere von Vitamin E in Bezug auf ihr Vorkommen in der Nahrung, ihre intrazelluläre Lokalisation bei der Aufnahme in Leberzellen und ihre Gewebeverteilung bei Säugetieren zu erweitern. Die potentielle Rolle des TTP bei der intrazellulären Verteilung und dem Transport im Organismus der nicht-alphaT-Kongenere war ein zweiter Schwerpunkt der aktuellen Untersuchungen.
Zu diesem Zweck wurden die Vitamin-E-Profile und -Gehalte in Ölen von drei Elaeis Guineensis, zwei Elaeis Oleifera und einem hybriden OxG-Palmenfrüchte-Genotyp aus Costa Rica ermittelt. Die Öle wurden entweder mechanisch mit einer Spindelpresse oder chemisch mit Hexan extrahiert. Die Vitamin-E-Profile in den Palmölen waren unabhängig vom Genotyp und Extraktionsverfahren ähnlich, alpha- und gamma-T3 waren die häufigsten Kongenere. alpha-T1 wurde in fünf der sechs Sorten gefunden. Die Extraktion mit Hexan ergab im Vergleich zur mechanischen Spindelpresse einen bis zu 2,5-fach höheren Gesamt-Vitamin E-Gehalt.
Aufgrund der aus den Ölen gewonnen Erkenntnisse bezüglich der Häufigkeit der vorkommenden nicht- alphaT-Kongenere, wurden die beiden Tocotrienole für weitere Studien auf ihre zelluläre Aufnahme und intrazelluläre Verteilung in kultivierten Leberzellen mit und ohne stabiler TTP-Expression ausgewählt und mit ihren jeweiligen Tocopherol-Gegenstücken verglichen. Nach Aufnahme der vier Kongenere waren diese hauptsächlich mit den Lysosomen, dem endoplasmatischen Retikulum und der Plasmamembran assoziiert. Zusammengefasst lassen diese Ergebnisse darauf schließen, dass weder die strukturellen Unterschiede, noch die Expression des TTP ausschlaggebende Faktoren für den intrazellulären Transport (Aufnahme und Verteilung) der Kongenere in kultivierten Leberzellen sind.
Abschließend wurde eine Tierstudie durchgeführt, um die Gewebeverteilung des alphaT1 bei Mäusen im Vergleich zu alphaT untersuchen. Ebenfalls wurde hierbei der Einfluss von TTP mit berücksichtigt. Wildtyp und TTP Knockout Mäuse erhielten 2 Wochen lang eine Standarddiät mit alphaT oder alphaT1. Die Konzentrationen wurden im Blut und verschiedenen Geweben bestimmt. Das alphaT1 konnte ausschließlich im Blut, nicht in den Geweben, gefunden werden. Der Verlust der TTP-Funktion bei Knockout Mäusen führte zu einer fast vollständigen Abnahme der alphaT-Konzentrationen in allen Geweben. Interessanterweise konnte alphaT1 noch im Blut nachgewiesen werden. Das Vorhandensein von alphaT1 im Blut von Mäusen mit und ohne TTP-Funktion deutet darauf hin, dass TTP für die Sekretion von alphaT1 in den systemischen Kreislauf nicht oder nur eingeschränkt erforderlich sein könnte.
Aufgrund des höheren Wissenstandes über alphaT im Vergleich zu den nicht-alphaT-Kongeneren, sind auf Grundlage der vorliegenden Arbeit neue Möglichkeiten für die weitere Erforschung der biologischen Aktivitäten und den daraus resultierenden gesundheitlichen Vorteile der nicht-alphaT-Kongenere entstanden.

Cosmetic products are important consumer goods in the "non-food" sector, which should not have negative effects of the human health. Critical compounds, however, can be formed by the oxidation of an unsaturated organic compound.Thereby formed oxidation products with potentially adverse properties are well known from the food sector. As the oxidation of cosmetic ingredients, however, has less been studied, the oxidation of selected cosmetic ingredients with respect to the formation of potentially critical compounds was investigated within the framework of this thesis.The oxidation of cholesterol to various cholesterol oxidation products (COPs) was investigated in a first step.COPs are known from the food sector and are suspected of causing certain diseases such as arteriosclerosis.Cosmetic products have not yet been tested for COPs, although a versatile ingredient used only in cosmetic products, lanolin, contains above-average levels of the cholesterol, which is the precursor.Total COPs contents in cosmetics containing lanolin, namely lip care products, fat creams and ointments for nursing women were in the low percent range (up to 3 %) and were thus several orders of magnitudes higher than the contents found in food.The oxidation of fragrances was studied in the second part of this work.The subject is not new as the oxidation of terpenes to contact allergens has been studied in earlier studies. The oxidation of other fragrances was hardly investigated. In order to extend our knowledge in this field, the oxidation of a synthetic fragrance frequently used in perfumes, octahydro tetramethyl naphthalenyl ethanone (OTNE) was studied. Obtained results indicated that peroxides of OTNE were formed during oxidation.It was found out that the OTNE oxidation even occurs, when perfumes are stored indoors under normal temperature and light conditions. An in-vivo test showed that OTNE oxidation can be expected on the skin after application of a perfume.
Kosmetische Produkte sind wichtige Konsumgüter des Non-Food Sektors, die keine negativen Effekte auf die Gesundheit des Mensches haben soll. Kritische Verbindungen können jedoch auch durch Folgereaktionen aus kosmetischen Inhaltsstoffen gebildet werden. Ein Beispiel für eine Folgereaktion ist die Oxidation einer ungesättigten organischen Verbindung.Dabei gebildete Oxidationsprodukte mit potentiell gesundheitsgefährdenden Eigenschaften sind aus dem Lebensmittelbereich bekannt. Da die Oxidation von kosmetischen Inhaltsstoffen weniger untersucht ist, wurde im Rahmen dieser Arbeit die Oxidation von ausgewählten kosmetischen Inhaltsstoffen im Hinblick auf die Bildung potentiell kritischer Verbindungen betrachtet.Im ersten Schritt wurde die Oxidation von Cholesterin zu verschiedenen Cholesterinoxidationsprodukten (COPs) untersucht. COPs sind aus dem Lebensmittelbereich bekannt und stehen im Verdacht, bestimmte Krankheiten wie Arteriosklerose hervorzurufen. Kosmetische Mittel wurden bisher nicht auf COPs untersucht, obwohl ein nur in kosmetischen Produkten vielseitig eingesetzter Inhaltsstoff, Lanolin, überdurchschnittlich hohe Gehalte am Vorläufer Cholesterin enthält. Gesamt COPs Gehalte in kosmetischen Mitteln mit Lanolin, nämlich Lippenpflegeprodukte, Fettcremen und Salben für stillende Frauen, waren im niedrigen Prozentbereich (bis 3 %) und lagen damit um einige Größenordnungen über den in Lebensmitteln gefundenen Gehalten.Im zweiten Teil der Arbeit wurde die Oxidation von Duftstoffen untersucht. Frühere Studien thematisierten die Oxidation von Terpenen zu Kontaktallergenen. Die Oxidation anderer Duftstoffe wurde kaum untersucht. Um die Kenntnis in diesem Bereich zu erweitern, wurde die Oxidation eines häufig in Parfums eingesetzten, synthetischen Duftstoffes, Octahydrotetramethylnaphthalenylethanon (OTNE), untersucht.Erhaltene Ergebnisse deuten darauf hin, dass bei der Oxidation Peroxide von OTNE gebildet wurden.Dass die OTNE Oxidation bereits bei Lagerung der Parfums im Innenraum unter normalen Temperatur- und Licht-Bedingungen erfolgt, wurde durch einen Lagerungstest herausgefunden. Ein in-vivo Test zeigte zudem, dass mit der OTNE Oxidation auf der Haut nach Anwendung eines Parfums zu rechnen ist.

The protection of the aquatic environment and the supply of clean drinking water to people all over the world are central challenges of our time. Monitoring of the aquatic environment and the input of anthropogenic trace substances into it is therefore very important. However, since aquatic environmental samples often consist of complex substance mixtures, their characterization and evaluation is very demanding. By using generic target analysis methods, selected known anthropogenic trace substances can be detected and quantified very sensitively. For the detection of previously unknown substances, non-target analysis methods have been increasingly used in recent years. However, these methods do not provide information on the relevance of the anthropogenic trace substances occurring in water. In this context, especially all those trace substances are regarded as relevant from which a harmful effect on humans or water organisms is to be expected. For the detection of such effective substances, effect-directed analysis (EDA) can be used.
In EDA, a bioassay is combined with a fractionation method and subsequent chemical analysis, the aim being to identify the bioactive substance. The separation method used in this work is high-performance thin-layer chromatography (HPTLC). After chromatography, the bioassay is performed directly on the HPTLC plate. If an effective zone appears in the bioassay, a prioritization strategy is used to clarify the identity of the substance.
Due to the complex aquatic samples, a large number of different substances in a zone must still be expected despite the applied HPTLC separation, which makes it difficult to identify the effective substance. Therefore, a strategy to simplify the identification of effective substances should be developed. The aim was to reduce the complexity by multidimensional separation in such a way that chemical analysis can be used to prioritize to a few candidates in the effective fraction. In the first part of the work, a selective two-dimensional HPTLC separation was developed to reduce the number of substances in a bioactive zone. After the first separation dimension (1D) the acetylcholinesterase inhibition assay (AChE assay) was performed and afterwards only the effective zones were extracted from the HPTLC plate. The selected effective zones were separated in a second separation dimension (2D) and the bioassay was performed again. With this 2D separation, the peak capacity could be increased by a factor of 7 compared to a 1D HPTLC gradient development. If real water samples are examined for their effects, an additional structural elucidation must be carried out to clearly identify the unknown bioactive substances. In this work, the developed 2D EDA was therefore connected to a high-performance liquid chromatography (HPLC) with high-resolution mass spectrometry (HRMS) and a non-target screening (NTS) was performed. Using three water samples(drinking water, surface water and purified sewage water) spiked with six effective substances, it was shown that the developed strategy is suitable for the identification of effective substances and that these can be recovered despite repeated extraction. When applying the developed methodology to real samples, it was also possible to assign and quantify the detected effect in several waters to the substance lumichrome and to linear alkylbenzene sulfonates.
Genotoxicity is a crucial endpoint for the effect assessment of water samples. However, this endpoint with metabolic activation cannot yet be performed directly on the HPTLC plate. Since many of the genotoxic substances have an indirect genotoxic effect, i.e. they only acquire their activity after metabolic activation; this endpoint was investigated in the present work with the umu assay in the microtiter plate. However, separation with HPTLC, subsequent extraction of effective zones and non-target analysis of the extracts, should also be performed for this assay. Therefore the umu assay in the microtiter plate was integrated into the existing EDA-with-HPTLC concept. In laboratory experiments, sodium hypochlorite was added to the drug metformin in order to simulate the behavior of the substance during water treatment (chlorination). The metformin sample treated with hypochlorite was examined with the umu assay and a genotoxic effect was detected. After HPTLC separation of the chlorinated sample, zones were extracted over the entire retardation range. When the extracted zones were examined with the umu assay, the genotoxic effect could be clearly assigned to one fraction. Using high-resolution mass spectrometry, the genotoxic effect could be assigned to an already known transformation product of metformin. The HPTLC separation and extraction of the zones from the plate led to a reduction of the possible effective candidate masses by a factor of 10 and thus to a clear prioritization in HRMS analysis.
Der Schutz der aquatischen Umwelt und die Versorgung der Menschen auf der ganzen Welt mit sauberem Trinkwasser sind zentrale Herausforderungen unserer Zeit. Daher ist die Überwachung der aquatischen Umwelt und des Eintrags von anthropogenen Spurenstoffen in diese von wichtiger Bedeutung. Durch den Einsatz klassischer Target-Analysemethoden können ausgewählte, bekannte anthropogene Spurenstoffe sehr empfindlich detektiert werden. Für die Detektion bislang unbekannter Substanzen werden Non-Target-Analysemethoden eingesetzt. Allerdings liefern diese Methoden keine Aussagen zur Relevanz der im Wasser vorkommenden anthropogenen Spurenstoffe. Als relevant gelten in diesem Zusammenhang all jene Spurenstoffe, von denen eine schädliche Wirkung auf den Menschen oder im Wasser lebende Organismen zu erwarten ist. Zur Detektion wirkender Substanzen kann die Wirkungsbezogene Analytik (WBA) verwendet werden. Bei der WBA wird ein Bioassay mit einer Fraktionierungsmethode und anschließender chemischer Analyse kombiniert, wobei das Ziel die Identifizierung der bioaktiven Substanz ist. Als Fraktionierungsmethode wurde in dieser Arbeit die Hochleistungsdünnschichtchromatographie (HPTLC) angewandt. Nach der Chromatographie wird der Bioassay direkt auf der HPTLC-Platte durchgeführt. Tritt im Bioassay eine wirkende Zone auf, so soll mithilfe einer Priorisierungsstrategie die Identität der Substanz aufgeklärt werden.
Aufgrund der komplexen aquatischen Proben muss trotz der angewandten HPTLC-Trennung in einer wirkenden Zone noch immer mit einer Vielzahl verschiedener Substanzen gerechnet werden, die eine Identifizierung der wirkenden Substanz erschweren. Es sollte aus diesem Grund eine Strategie zur Vereinfachung der Identifizierung von wirkenden Substanzen entwickelt werden. Ziel war es, die Komplexität durch mehrdimensionale Trennung so zu verringern, dass mithilfe der chemischen Analyse auf wenige Kandidaten in der wirkenden Zone priorisiert werden kann. Im ersten Teil der Arbeit wurde zur Reduktion der Anzahl von Substanzen in einer bioaktiven Zone eine selektive zweidimensionale HPTLC-Trennung entwickelt. Nach der ersten Trenndimension (1D) wurde der Acetylcholinesterase-Hemmtest (AChE-Assay) durchgeführt und anschließend die wirkenden Zonen von der HPTLC-Platte extrahiert. Die ausgewählten wirkenden Zonen wurden in einer zweiten Trenndimension (2D) nochmals aufgetrennt und abermals der Bioassay durchgeführt. Die Peakkapazität konnte durch diese 2D-Trennung im Vergleich zu einer 1D-HPTLC Gradientenentwicklung um den Faktor 7 gesteigert werden.
Wenn reale Wasserproben wirkungsbezogenen untersucht werden, muss für eine eindeutige Identifizierung der unbekannten bioaktiven Substanzen zusätzlich eine Strukturaufklärung durchgeführt werden. In dieser Arbeit wurde deshalb die entwickelte 2D-WBA an eine Hochleistungsflüssigkeitschromatographie (HPLC) mit hochauflösender Massenspektrometrie (HRMS) angebunden und ein Non-Target-Screening (NTS) durchgeführt. Am Beispiel dreier mit sechs wirkenden Substanzen dotierter Wasserproben konnte gezeigt werden, dass sich die entwickelte Strategie für die Identifizierung wirkender Substanzen eignet und dass diese trotz mehrmaliger Extraktion wiedergefunden werden können. Bei der Anwendung der entwickelten Methodik auf Realproben gelang es, in mehreren Wässern die detektierte Wirkung der Substanz Lumichrome sowie linearen Alkylbenzolsulfonaten zuzuordnen.
Für die Beurteilung der Wirkungen von Wasserproben ist die Gentoxizität ein entscheidender Endpunkt. Allerdings kann dieser Endpunkt mit metabolischer Aktivierung bislang nicht direkt auf der HPTLC-Platte durchgeführt werden. Da jedoch viele der gentoxischen Substanzen indirekt gentoxisch wirken, wurde dieser Endpunkt mit dem umu-Assay in der Mikrotiterplatte untersucht. Jedoch sollten auch für diesen Assay eine Fraktionierung mit der HPTLC, eine anschließende Extraktion wirkender Zonen und Non-Target-Analyse der Extrakte erfolgen. Dazu wurde der umu-Assay in der Mikrotiterplatte in das bestehende WBA-mit-HPTLC-Konzept integriert.
Im Laborexperiment wurde das Arzneimittel Metformin mit Natriumhypochlorit versetzt, um damit das Verhalten der Substanz während der Wasseraufbereitung nachzustellen. Die mit Hypochlorit umgesetzte Metforminprobe wurde mit dem umu-Assay untersucht, wobei eine gentoxische Wirkung detektiert werden konnte. Nach der HPTLC-Trennung der gechlorten Probe wurden anschließend Zonen über den gesamten Retardationsbereich extrahiert. Bei der Untersuchung der extrahierten Zonen mit dem umu-Assay konnte der gentoxische Effekt eindeutig einer Fraktion zugeordnet werden. Mittels hochauflösender Massenspektrometrie gelang es, den gentoxischen Effekt einem bereits beschriebenen Transformationsprodukt von Metformin nachzuweisen. Die HPTLC-Trennung und Extraktion der Zonen von der Platte führte zu einer Reduktion der möglichen wirkenden Kandidatenmassen um den Faktor 10 und somit zu einer deutlichen Priorisierung bei der HRMS-Analyse.

Diese Doktorarbeit befasst sich mit zwei wichtigen astrobiologischen Aspekten von Metalloporphyrinen. Erstens mit der möglichen abiotischen Entstehung von Metalloporphyrinen unter präbiotisch plausiblen Bedingungen und zweitens mit Metalloporphyrinen als mögliche molekulare Biosignaturen im Zusammenhang mit der Suche nach außerirdischem Leben. Diese zweigleisige Herangehensweise ermöglichte es, einen umfassenderen Überblick über Metalloporphyrine im astrobiologischen Kontext zu gewinnen.
In den heute bekannten lebenden Organismen kommen Metallkomplexe von Porphyrinen und Porphyrinoiden ubiquitär vor; freie Porphyrinbasen spielen dagegen kaum eine Rolle. Zu den bekanntesten Beispielen zählen die Chlorophylle (Mg-Komplexe), die entscheidend an der Photosynthese beteiligt sind, und die Häm-Gruppe, z. B. als Cofaktor der Cytochrome, welche eine bedeutende Rolle bei der Zellatmung spielen. Evolutiv scheinen Porphyrinoid-Cofaktoren sehr alt zu sein, und tatsächlich wurden 1,1 Milliarden Jahre alte Geoporphyrine in Sedimentgestein gefunden. Vorfahren der heute bekannten Porphyrinoid-Cofaktoren, einfache Metalloporphyrine, könnten schon in den ersten Lebewesen oder noch früher in abiotischen Protometabolismen vorgekommen sein. Zudem legt die weite Verbreitung von Porphyrinoid-Cofaktoren in den bekannten Organismen sowie ihre Beteiligung an grundlegenden biologischen Funktionen nahe, dass auch in möglichen Lebensformen außerhalb der Erde Metalloporphyrinoide vorkommen könnten. Somit könnten Metalloporphyrinoide als chemische Biosignaturen für die Suche nach Leben auf Mars, Europa, Enceladus und darüber hinaus geeignet sein.
Das erste Ziel dieser Arbeit war es, zu untersuchen, ob eine Komplexbildung mit ausgewählten Metallen (Fe, Mg, Co, Ni und Cu) unter präbiotisch plausiblen Bedingungen möglich ist. Ausgehend von einer zuvor von unserer Arbeitsgruppe beschriebenen präbiotischen Synthese von Octaalkylporphyrinen an simulierten urzeitlichen Vulkanküsten sollte der Einfluss von Nass-Trocken-Zyklen (NTZ) auf Octaethylporphyrin (OEP) und ausgewählte Metallquellen untersucht werden. Hierzu musste zunächst eine neuartige, automatisierte Apparatur entwickelt werden, die die Simulation der Bedingungen auf der frühen Erde, speziell an urzeitlichen Vulkanküsten, ermöglichte. Insbesondere musste diese Apparatur einen strikten Ausschluss von Luftsauerstoff sicherstellen und fluktuierende Wechsel zwischen Nass- und Trockenphasen ermöglichen.
Zunächst wurden Experimente durchgeführt, um die neue Apparatur zu testen. Dabei konnte gezeigt werden, dass die Apparatur über einen längeren Zeitraum vollständig automatisch und zuverlässig arbeitete. In weiteren Versuchen wurde geprüft, ob oxidationsempfindliche Sub-stanzen in der Apparatur gehandhabt werden können. Hierbei konnte gezeigt werden, dass in der Tat ein strikter Ausschluss von Sauerstoff möglich ist. Damit erwies sich die Apparatur als geeignet für die Untersuchung der Bildung von Metall¬komplexen aus OEP und diversen Metallquellen unter dem Einfluss von wechselnden NTZ. Als Metallquellen wurden unter anderem Metall(II)-chloride, Metallsulfide, Basalt und Eisenmeteorite eingesetzt. Hierbei lag der Fokus auf Eisenquellen. Tatsächlich konnte gezeigt werden, dass sich mit den ent¬sprechenden Metallquellen Eisen-, Magnesium-, Cobalt
Nickel- und Kupfer-Komplexe in einer ungewöhnlichen Reaktion bildeten (vollständig wasserunlösliches OEP reagierte mit teilweise ebenfalls unlöslichen Metallquellen), wobei sich NTZ als essenziell erwiesen. In Süßwasser konnten Ausbeuten zwischen 20 und 78 % (bezogen auf das Porphyrin) erhalten werden. Außerdem wurde der Einfluss von künstlichem Meerwasser und niedrigen pH-Werten auf die Komplexbildung untersucht.
Im zweiten Teil der Arbeit wurde der Frage nach der Stabilität von Metalloporphyrinen modellhaft anhand der Verbindung Chlorido(octaethylporphyrinato)eisen(III), [FeCl(oep)], nachgegangen. Für die durchgeführten Experimente wurden potentiell destruktive Bedingungen ausgewählt, die für Metalloporphyrine als mögliche chemische Biosignaturen relevant sind, aber auch Bedingungen, wie sie vermutlich an urzeitlichen Vulkanküsten herrschten. In umfangreichen Versuchsreihen konnte unter anderem gezeigt werden, dass (a) der Eisen-OEP-Kern über einen pH-Bereich von 0,0 bis 13,5 stabil ist, (b) [FeCl(oep)] bis ca. 250 °C in inerter Atmosphäre stabil ist, (c) eine Salzmatrix [FeCl(oep)] vor Röntgenstrahlung schützt, aber nicht vor Eisen-Partikelstrahlung, und (d) Hypochlorit, Wasserstoffperoxid, Chlorat und Salpetersäure [FeCl(oep)] oxidativ zersetzen (Oxidationswirkung in absteigender Reihenfolge). Perchlorat, das häufig im Zusammenhang mit seinem Vorkommen im Mars-Regolith genannt wird, zeigte dagegen keine Oxidationswirkung. Schließlich wird in der vorgelegten Arbeit die Bedeutung der Metalloporphyrine als potentielle Biosignaturen diskutiert, speziell vor dem Hintergrund ihrer Stabilität und möglicher abiotischer Synthesewege für diese Verbindungen.
This doctoral thesis addresses two important astrobiological aspects of metalloporphyrins. First, the possible abiotic origin of metalloporphyrins under prebiotically plausible conditions, and second, metalloporphyrins as possible molecular biosignatures in the context of the search for extraterrestrial life. This two-pronged approach allowed us to obtain a more comprehensive overview of metalloporphyrins in an astrobiological context.
In presently known living organisms, metal complexes of porphyrins and porphyrinoids occur ubiquitously, whereas free base porphyrins scarcely matter. Among the best known examples are chlorophylls (magnesium complexes), which are crucially involved in photosynthesis, and the heme group, for example as a cofactor of cytochromes, which play an important role in cel¬lular respiration. In terms of evolution, porphyrinoid cofactors appear to be very old, and indeed 1.1 billion-year-old geoporphyrins have been found in sedimentary rocks. Ancestors of current porphyrinoid cofactors, simple metalloporphyrins, may have been present in the first organisms or even earlier in abiotic protometabolisms. Furthermore, the wide distribution of porphyrinoid cofactors among known organisms, as well as their participation in basic biological functions, suggests that metalloporphyrinoids may also be present in potential life forms beyond Earth. Thus, metalloporphyrinoids could be useful as chemical biosignatures in the search for life on Mars, Europa, Enceladus and beyond.
The first objective of this work was to investigate whether complex formation with selected metals (Fe, Mg, Co, Ni, and Cu) is possible under prebiotically plausible conditions. Based on a prebiotic synthesis of octaalkylporphyrins at simulated primordial volcanic coasts, which has been previously described by our group, the influence of wet-dry cycles on octaethylporphyrin (OEP) and selected metal sources was to be studied. For this purpose, first a novel, automated apparatus had to be developed that allowed the simulation of conditions on the early Earth, especially at primordial volcanic coasts. In particular, this apparatus had to ensure strict exclu-sion of atmospheric oxygen and allow fluctuating changes between wet and dry phases (simu-lation of tides or rainfall).
Initially, experiments were conducted to test the new apparatus. It was shown that the appa-ratus worked completely automatically and reliably over a longer period of time. In further experiments, it was tested whether oxidation-sensitive substances can be handled in the apparatus. Indeed, it was shown that it is possible to strictly exclude oxygen. Thus, the apparatus proved to be suitable for investigating the formation of metal complexes from OEP and various metal sources under the influence of alternating wet-dry cycles. Metal sources used included metal(II) chlorides, metal sulfides, basalt, and iron meteorites. The focus was on iron sources. Indeed, it was shown that iron, magnesium, cobalt, nickel and copper complexes formed with the corresponding metal sources in an unusual reaction (completely water-insoluble OEP reacted with partly also insoluble metal sources), whereby wet-dry cycles turned out to be essential. Yields ranging from 20 to 78% (relative to the porphyrin) were obtained in fresh water. In addition, the influence of artificial seawater and low pH values on complex formation was investigated.
In the second part of the thesis, the stability of metalloporphyrins was investigated using the compound chlorido(octaethylporphyrinato)iron(III), [FeCl(oep)], as a model. Potentially de-structive conditions relevant to metalloporphyrins as possible chemical biosignatures were selected for the experiments performed, as well as conditions that presumably prevailed on primordial volcanic coasts. Extensive series of experiments demonstrated, among other things, that (a) the iron OEP core is stable over a pH range of 0.0 to 13.5, (b) [FeCl(oep)] is stable up to ca. 250 °C in an inert atmosphere, (c) a salt matrix protects [FeCl(oep)] from X-ray radiation but not from iron particle radiation, and (d) hypochlorite, hydrogen peroxide, chlorate, and nitric acid oxidatively decompose [FeCl(oep)] (oxidation power in descending order). On the other hand, perchlorate, which is often mentioned in connection with its occurrence in the Martian regolith, did not show any oxidation effect. Finally, the importance of metalloporphyrins as potential biosignatures is discussed in the presented work, particularly in the light of their stability and possible abiotic synthetic pathways for these compounds.

Heterocyclen sind Grundgerüste zahlreicher Arzneimittel und daher von hoher Bedeutung in der Medizinischen Chemie. Infolgedessen besteht naturgemäß eine hohe Nachfrage nach Methoden zum Aufbau von Heterocyclen. Die Anforderungen an neue Synthesemethoden für Heterocyclen sind mittlerweile sehr hoch, da sie nicht nur effizient und selektiv, sondern darüber hinaus auch nachhaltig sein müssen. Diese Anforderungen können sowohl durch übergangsmetallfreie als auch übergangsmetallkatalysierte Reaktionen erfüllt werden. So gelingt die übergangsmetallfreie Darstellung einer Vielzahl unterschiedlicher Heterocyclen durch radikalische, kationische und anionische Cyclisierungen sowie durch pericyclische Reaktionen. Seit kurzem nimmt die Bedeutung von elektro- und photochemischen Methoden in der Heterocyclen-Synthese sehr stark zu. Bei der übergangsmetallkatalysierten Heterocyclen-Synthese spielen vor allem Pd- und Cu-Katalysatoren eine herausragende Rolle. Zum Pd-katalysierten Aufbau von N-Heterocyclen ist insbesondere die intramolekulare Buchwald-Hartwig-Aminierung hervorzuheben. Mittlerweile weiß man, dass sich viele Pd-katalysierte Reaktionen auch mit Cu-Katalysatoren durchführen lassen. Vor dem Hintergrund, dass Cu-Katalysatoren aufgrund der höheren Häufigkeit von Cu wesentlich preisgünstiger sind und dass man zur Durchführung Cu-katalysierter Reaktionen meistens auf teure Liganden verzichten kann, ist ihre sehr große Bedeutung in der Heterocyclen-Synthese leicht nachzuvollziehen. Beispielsweise lassen sich viele N-Heterocyclen durch intramolekulare Ullmann-Reaktionen mit hervorragenden Ausbeuten und hohen Selektivtäten darstellen. Hierbei spielen bisfunktionalisierte Substrate mit zwei Zentren unterschiedlicher Reaktivität eine tragende Rolle. Ziel der vorliegenden Arbeit war es, neue effiziente und hochselektive Synthesemethoden zum Aufbau relevanter N- bzw. O-Heterocyclen zu entwickeln. Dabei sollten insbesondere Cu-katalysierte Reaktionen mit bisfunktionalisierten Substraten entwickelt werden. Es sollte zudem geprüft werden, ob sich die entsprechenden Umsetzungen auch in Abwesenheit von Übergangsmetall-Katalysatoren durchführen lassen. Benzodioxine und 2,3-Dihydrobenzodioxine weisen viele interessante biologische Eigenschaften auf, jedoch sind die bislang für ihre Synthese zur Verfügung stehenden Möglichkeiten überschaubar. Darum beschäftigt sich der erste Teil dieser Dissertation mit der Entwicklung einer neuen Methode zum diastereospezifischen Aufbau von (Z)-2-Aryliden-2,3-dihydrobenzodioxinen (Z)-80 (Schema 48) durch Umsetzung von 3-arylsubstituierten (Z)-1,2-Dibromarylpropenen (Z)-82 mit Catecholen 83. Während das Modellsubstrat (Z)-82a (R1 = Ph) durch Reduktion und nachfolgende Bromierung von a-Bromzimtaldehyd dargestellt werden kann, wurden die übrigen Substrate (Z)-82 in drei Stufen aus den entsprechenden Benzaldehyden hergestellt. Die anschließende Optimierung der Modellreaktion unter verschiedenen Reaktionsbedingungen ergab, dass sich die besten Ergebnisse unter übergangsmetallfreien Bedingungen erzielen ließen. Die höchste Ausbeute an (Z)-80a (R1 = Ph, R2 = H) erhielt man, wenn man 1 Äquivalent (Z)-82a mit zwei Äquivalenten 83a (R2 = H) in Gegenwart von vier Äquivalenten Cs2CO3 in DMF 18 h bei 140 °C umsetzte. Bemerkenswert ist, dass diese übergangsmetallfreie Domino-Reaktion, die aus einer intermolekularen O-Allylierung und einer nachfolgenden intramolekularen O-Vinylierung besteht, hochdiastereospezifischen verläuft: bei Einsatz von (Z)-1,2-Dibrom-3-phenyl-2-propen [(Z)-82a] entsteht ausschließlich (Z)-2-Benzyliden-2,3-dihydrobenzodioxin [(Z)-80a]. Diese hohe Diastereospezifität wurde auch bei den Umsetzungen aller anderen Substrate (Z)-82 beobachtet. Die 2-Aryliden-2,3-dihydrobenzodioxine (Z)-80 konnten mit Ausbeuten von bis zu 89% erhalten werden. Die Methode toleriert sowohl unterschiedliche Substituenten am Aromaten von (Z)-82 als auch unterschiedlich disubstituierte Catechole 83. DFT-Kalkulationen, die in Kooperation mit Prof. Bharatham, NIPER Nagar (Mohali), entstanden, legen nahe, dass die intramolekulare O-Vinylierung nicht über eine Alkin- sondern über eine Alkenzwischenstufe verläuft. Die diastereospezifische Umsetzung des E-konfigurierten Substrats (E)-82a (R1 = Ph) zum entsprechenden (E)-2-Benzyliden-2,3-dihydrobenzodioxin [(E)-80a] unterstützt diese Annahme. Der zweite Teil dieser Arbeit beschäftigte sich mit der intramolekularen Cu(I)-katalysierten Cyclisierung von o-Halobenzylidenguanylhydrazon-Salzen (E)-86 zum direkten Aufbau von N-1 unsubstituierten 1H-Indazolen 84 (Schema 49). Die Synthese von Indazolen dieses Typs ist von besonderem Interesse für die Medizinische Chemie, weil sie das Grundgerüst einiger wichtiger Krebsmedikamente bilden. Die Substrate (E)-86 wurden durch Kondensation von o-Halobenzaldehyden mit Aminoguanidin-Hydrochlorid mit Ausbeuten von bis zu 90% dargestellt. Die anschließende Cyclisierung unter Verwendung von 10 mol% CuI, 30 mol% DMEDA und 0.5 Äquivalenten Cs2CO3 lieferte die 1H-Indazole 84 in Ausbeuten von bis zu 75%. Die Umsetzungen wurden bei 120 °C in DMF für 5 h in einem verschlossenen Glasröhrchen durchgeführt. Die Methode toleriert eine ganze Reihe von Substituenten am Aromaten der Substrate. Aufgrund von DFT-Kalkulationen, die in Kooperation mit Prof. Bharatham, NIPER Nagar (Mohali), entstanden, kann man davon ausgehen, dass es zunächst zu einer E/Z-Isomerisierung des Substrats 86 kommt, an die sich eine Metall-Komplexierung mit nachfolgender C,N-Bindungsbildung anschließt. Die abschließende Hydrolyse des 1H-Indazol-1-carboximidamids liefert das N-1 unsubstituierte 1H-Indazol 84.
Heterocycles are the backbones of numerous drugs and are therefore of great importance in medicinal chemistry. As a result, there is an inevitably high demand for methods to synthesize heterocycles. The requirements for new methods for the synthesis of heterocycles are nowadays very high, as they must not only be efficient and selective, but also sustainable. These prerequisites can be met by both transition metal-free and transition metal-catalyzed reactions. Thus, the transition metal-free preparation of a variety of different heterocycles can be achieved by radical, cationic and anionic cyclizations as well as by pericyclic reactions. Recently, the importance of electrochemical and photochemical methods in heterocyclic synthesis has been increasing very rapidly. In transition metal-catalyzed heterocycle synthesis, Pd- and Cu-catalysts in particular play a prominent role. For the Pd-catalyzed assembly of N-heterocycles, the intramolecular Buchwald-Hartwig amination is especially noteworthy. It is now known that many Pd-catalyzed reactions can also be carried out with Cu-catalysts. In view of the fact that Cu-catalysts are much cheaper due to the higher abundance of Cu, and that expensive ligands can usually be omitted to carry out Cu-catalyzed reactions, their enormous importance in heterocyclic synthesis is easy to understand. For example, many N-heterocycles can be prepared by intramolecular Ullmann reactions with excellent yields and high selectivities. Here, bisfunctionalized substrates with two centers of different reactivity play a major role.
The aim of the present work was to develop new efficient and highly selective synthetic methods for the construction of relevant N- or O-heterocycles. In particular, Cu-catalyzed reactions with bisfunctionalized substrates were to be developed. The investigation included determining whether the corresponding reactions can also be carried out in the absence of transition metal catalysts.
Benzodioxines and 2,3-dihydrobenzodioxines exhibit many interesting biological properties, but the possibilities available nowadays for their synthesis are limited. Therefore, the first part of this dissertation deals with the development of a new method for the diastereospecific construction of (Z)-2-arylidene-2,3-dihydrobenzodioxines (Z)-80 (Scheme 50) by reacting 3-aryl-substituted (Z)-1,2-dibromoarylpropenes (Z)-82 with catechols 83. While the model substrate (Z)-82a (R1 = Ph) can be prepared by reduction and subsequent bromination of a-bromocinnamaldehyde, the remaining substrates (Z)-82 were prepared in three steps from the corresponding benzaldehydes. Subsequent optimization of the model reaction under a wide variety of reaction conditions showed that the best results could be obtained under transition metal-free conditions.
The highest yield of (Z)-80a was obtained when 1 equivalent of (Z)-82a (R1 = Ph, R2 = H) was reacted with two equivalents of 83a (R2 = H) in the presence of four equivalents of Cs2CO3 in DMF for 18 h at 140 °C. Remarkably, this transition metal-free domino reaction, which consists of an intermolecular O-allylation followed by an intramolecular O-vinylation, is highly diastereospecific: the use of (Z)-1,2-dibromo-3-phenyl-2-propene [(Z)-82a] exclusively delivers (Z)-2-benzylidene-2,3-dihydrobenzodioxines [(Z)-80a]. This high diastereospecifity was also observed in the reactions of all other substrates (Z)-82. The 2-arylidene-2,3-dihydrobenzodioxins (Z)-80 were obtained in yields up to 89%. This method tolerates different substituents on the aromatic moiety of (Z)-82 as well as different disubstituted catechols 83. DFT calculations conducted in collaboration with Prof. Bharatham, NIPER Nagar (Mohali), suggest that the intramolecular O-vinylation proceeds via an alkene intermediate rather than an alkyne intermediate. The diastereoselective conversion of the E-configured substrate (E)-82a (R1 = Ph) to the corresponding (E)-2-benzylidene-2,3-dihydrobenzodioxine [(E)-80a] supports this assumption. The second part of this work is devoted to the intramolecular Cu(I)-catalyzed cyclization of o-haloarylideneguanylhydrazone salts (E)-86 for the direct construction of N-1 unsubstituted 1H-indazoles 84 (Scheme 51). The synthesis of indazoles of this type is of particular interest to medicinal chemistry because they form the backbone of some important anticancer drugs. Substrates (E)-86 were prepared by condensation of o-halobenzaldehydes with aminoguanidine hydrochloride in yields up to 90%. Subsequent cyclization using 10 mol% CuI, 30 mol% DMEDA and 0.5 equivalents of Cs2CO3 afforded the 1H-indazoles 84 in yields up to 75%. The reactions were carried out at 120 °C in DMF for 5 h in a sealed glass tube. The method tolerated a full range of substituents on the aromatic moiety of the substrates. Based on DFT calculations done in collaboration with Prof. Bharatham, NIPER Nagar (Mohali), it is reasonable to assume that E/Z isomerization of substrate 86 occurs first, followed by metal complexation with subsequent C,N bond formation. The final hydrolysis of the 1H-indazole-1-carboximidamide yields the N-1 unsubstituted 1H-indazole 84.

Essential oils belong to secondary plant metabolites, with terpenoids and phenylpropanoids being among the main constituents in terms of quantity. Due to their lipophilic character and high volatility, they are mainly obtained by steam distillation. Citrus essential oils (agrumen oils) are an exception
since they are usually extracted from the peels by means of pressing, whereby less volatile components such as coumarins and furocoumarins are also introduced. Due to their odor and taste-giving properties, essential oils are used in the food, beverage, and cosmetics industries. In addition, due to a wide range of pharmacological properties, they are used in phytotherapy as well as in aromatherapy. However, most essential oils are highly susceptible to oxidation, polymerization, dehydrogenation, and isomerization reactions in the presence of atmospheric oxygen, light, and at high temperatures. The resulting organoleptic changes usually lead to a significant quality reduction. The formation of terpene hydroperoxides is another problem, as these are suspected of causing intolerances such as redness and itching in 1-3% of the European population upon contact with the skin. The detection of these chemical changes forms an integral part of quality control and can be prevented as far as possible by suitable production, transport, and storage strategies. Due to their volatility, essential oils are mainly analyzed by gas chromatography. However, due to their instability, the detection of hydroperoxides places considerable demands on common analytical methods. For this reason, a novel spectrophotometric method for the detection of peroxides and hydroperoxides in terpenes and essential oils was developed (paper 1). The oxidation of N-N-dimethyl-p-phenylenediamine by peroxides yielding an intensely red-colored cation (Wursters red) allowed colorimetric detection and quantitation of even smallest amounts (LOD: 0.5 ppm). The minimal sample amount of only a few milligrams, as well as simple and fast performance predestine this method for daily laboratory routine (paper 1). Among plant terpenoids, the monoterpene R-(+)-limonene is very widespread. Thus, it is not only found in citrus oils but also of in caraway oil, where its proportion amounts to almost 50%. To investigate the storage stability, R-(+)-limonene, S-(+)-carvone, different caraway oils, and the corresponding caraway seeds were stored in desiccators at 25 °C and 40 °C for eighteen months (paper 2). The samples were analyzed monthly by GC/MS and GC/FID, as well as HPLC/DAD-MS/MS. This showed that the comparison of seed, isolated essential oil, and pure substance, whichhad not been considered in storage studies so far, was of extraordinary importance. Here, both the plant matrix and the essential oil had a protective effect on individual terpenes and delayed their degradation (paper 2). Further, a clear difference between photo-oxidation and autoxidation was observed. Light-induced oxidation of terpenes primarily resulted in the formation of hydroperoxides, whereas autoxidation led to a variety of compounds such as alcohols, ketones, and epoxides. Thus, the secondary products can serve as specific markers for conclusions about the preload and quality of essential oils. In the study presented in paper 3, further photo-oxidation experiments were conducted with beta-pinene, R-(+)-limonene, and gamma-terpinene, with added furocoumarins. Furocoumarins can absorb UV-A light in the range of 320 380 nm and enter an energetically excited state. This energy difference between the ground state and excited state can be dissipated again by the emission of fluorescent and phosphorescent light. In this process, short-wave energy-rich UV light is converted into lower-energy visible light (bathochromic shift). For this reason, the UV light-induced degradation of the terpenes beta-pinene, R-(+)-limonene, and gamma-terpinene could be significantly reduced by adding 5% each of xanthotoxin, bergapten, bergaptol, and bergamottin. The effect of adding bergaptol was most pronounced in the photooxidation of gamma-terpinene (paper 3). Consequently, in citrus essential oils from which the natural furocoumarins had been previously removed, irradiation with UV light resulted in a strong degradation of the terpenes. This process could be markedly reduced by the re-addition of 5% of the previously removed plant-specific furocoumarins (paper 4). In summary, it can be concluded that not only the plant matrix and the essential oil as a multicomponent mixture but also potential interactions with other substances forming part of the essential oil such as furocoumarins may significantly slow down the oxidation of terpenoids.
Ätherische Öle gehören zu den sekundären Pflanzenstoffen, wobei Terpenoide und
Phenylpropanoide mengenmäßig zu den Hauptbestandteilen zählen. Aufgrund ihres
lipophilen Charakters und ihrer hohen Flüchtigkeit werden diese überwiegend mittels Wasserdampfdestillation gewonnen. Eine Ausnahme bilden ätherische Zitrusöle (Agrumenöle), die in der Regel mittels Pressverfahren aus den Schalen gewonnen werden, wobei auch weniger flüchtige Komponenten wie z.B. Cumarine und
Furocumarine in die Ölphase übergehen. Aufgrund ihrer geruchs- und geschmacksgebenden Eigenschaften finden ätherische Öle Anwendung in der Lebensmittel-, Getränke- und Kosmetikindustrie. Darüber hinaus werden sie aufgrund zahlreicher pharmakologischer Eigenschaften in der Phytotherapie sowie in der Aromatherapie eingesetzt. Allerdings sind die meisten ätherischen Öle in Gegenwart von Luftsauerstoff, Licht und bei erhöhten Temperaturen sehr anfällig für Oxidations-, Polymerisations-, Dehydrierungs- und Isomerisierungsreaktionen. Die damit einhergehenden organoleptischen Veränderungen führen meist zu einer deutlichen Qualitätsminderung. Ein weiteres Problem stellt die Bildung von
Terpenhydroperoxiden dar, da diese im Verdacht stehen, bei 1-3% der europäischen Bevölkerung bei Hautkontakt Unverträglichkeiten wie Rötungen und Juckreiz auszulösen. Die Erfassung derartiger chemischer Veränderungen ist fester Bestandteil der Qualitätskontrolle, und diese sind durch geeignete Herstellungs-, Transport- und Lagerbedingungen zu minimieren. Die Analytik der ätherischen Öle findet aufgrund ihrer Flüchtigkeit vorwiegend mittels Gaschromatographie statt. Die Detektion von Hydroperoxiden stellt aufgrund ihrer Instabilität jedoch erhebliche Anforderungen an die herkömmlichen Analysemethoden. Deshalb wurde in der in Paper 1 vorgestellten Studie eine neue spektrophotometrische Methode zur Detektion und Quantifizierung von Peroxiden und Hydroperoxiden in Terpenen und ätherischen Ölen entwickelt. Die Oxidation von N-N-Dimethyl-p-phenylendiamin durch Peroxide zu einem intensiv rot gefärbten Kation (Wursters Rot) ermöglichte die kolorimetrische Detektion und Quantifizierung selbst kleinster Mengen (LOD 0,5 ppm). Die geringe Probenmenge von nur wenigen Milligramm sowie die einfache und schnelle Durchführung prädestinieren diese Methode für die tägliche Laborroutine (Paper 1).
Unter den pflanzlichen Terpenoiden ist das Monoterpen R-(+)-Limonen sehr weit verbreitet. So findet es sich nicht nur in Zitrusölen, sondern auch mit einem Anteil von knapp 50% in Kümmelöl. Zur Untersuchung der Lagerstabilität wurden R-(+)-Limonen, S-(+)-Carvon, verschiedene Kümmelöle und die entsprechende Kümmelsaat achtzehn Monate lang bei 25 °C und 40 °C in Exsikkatoren gelagert (Paper 2). Mittels GC/MS und GC/FID sowie HPLC/DAD-MS/MS wurden die Proben monatlich analysiert. Hierbei zeigte sich, dass der bisher in Lagerstudien nicht berücksichtige Vergleich von Saat, ätherischem Öl und Reinstoff von außerordentlicher Bedeutung ist. Dabei wirkten sich sowohl die Pflanzenmatrix als auch das ätherische Öl schützend auf die einzelnen Terpene aus und verzögerten deren Abbau (Paper 2). Dabei zeigte sich zudem ein deutlicher Unterschied zwischen der Photooxidation und der hier ablaufenden Autoxidation. Bei der lichtinduzierten Oxidation von Terpenen werden primär Hydroperoxide gebildet, während die Autoxidation zu einer Vielzahl von Verbindungen wie Alkoholen, Ketonen und Epoxiden führt. So können die Folgeprodukte als spezifische Marker zur Bewertung der Vorbelastung und Qualität eines ätherischen Öles herangezogen werden. In der in Paper 3 dargestellten Studie wurden weitere Photooxidationsversuche mit beta-Pinen, R-(+)-Limonen und gamma-Terpinen durchgeführt, denen zuvor Furocumarine zudotiert wurden. Furocumarine können UVA Licht im Bereich 320 380 nm absorbieren und gehen dabei in einen energetisch angeregten Zustand über. Die Energiedifferenz zwischen Grundzustand und angeregtem Zustand kann durch Emission von Fluoreszenz- und Phosphoreszenzlicht dissipiert werden. Hierbei wird energiereiches kurzwelliges UV-Licht in energieärmeres, sichtbares Licht umgewandelt (bathochromer Shift). Aus diesem Grund konnte der UV-Licht induzierte Abbau der Terpene beta-Pinen, R-(+)-Limonen und gamma-Terpinen durch die Zugabe von je 5% Xanthotoxin, Bergapten, Bergaptol und Bergamottin erheblich reduziert werden. Der deutlichste Effekt wurde bei Zugabe von Bergaptol zu gamma-Terpinen beobachtet (Paper 3). Folgerichtig kam es bei ätherischen Zitrusölen, aus denen die natürlichen Furocumarine zuvor entfernt wurden, durch Bestrahlung mit UV-Licht zu einem starken Abbau der Terpene. Dieser Prozess konnte durch erneute Zugabe von 5% der zuvor entfernten pflanzenspezifischen Furocumarine deutlich verringert werden (Paper 4).
Zusammenfassend lässt sich feststellen, dass nicht nur die pflanzliche Matrix und das ätherische Öl als Mehrkomponentengemisch, sondern auch die Wechselwirkung mit anderen, im ätherischen Öle enthaltenen Substanzen wie Furocumarinen die Oxidation von Terpenoiden signifikant verlangsamen kann.
